| PART ⅠObjective:Critical roles of Protein Tyrosine Phosphotase Receptor type-O(PTPRO)and Toll-Like Receptor 4(TLR4)have been implicated in inflammation.However,little is known about their functional effects on Atherosclerosis(AS).We aim to study their potential function in AS.Methods:Oxidized Low-Density Lipoprotein(ox-LDL)induced AS models were constructed with PTPRO over-expressing RAW264.7 cells and PTPRO knockout macrophages.Cell apoptosis was assayed by flow cytometry and fatty accumulation was evaluated by oil red staining.The production of Reactive Oxygen Species(ROS),Superoxide Dismutase(SOD),Malondialdehyde(MDA),Triglyceride(TG),and Total Cholesterol(TC)were evaluated.Western blot was performed to detect the expression of Cluster of Differentiation 36(CD36),TLR4 and Nuclear Factor Kappa B(NF-κB).Results:PTPRO expression was promoted in a dose-dependent and time-dependent manner following ox-LDL challenging.In PTPRO-over-expressing cells,CD36 expression and the level of oil-red staining,TC and TG were increased,ROS production and MDA were improved,cell apoptosis was enhanced,but SOD was reduced.However,in PTPRO knockout cells,opposite results were found.TLR4 and NF-κB/p65 phosphorylation were significantly enhanced in PTPRO over-expressing cells,while significantly down-regulated in PTPRO knockout cells.Conclusion:PTPRO plays ital roles in AS via promoting ox-LDL induced oxidative stress and cell apoptosis through TLR4/NF-κB pathway.PART ⅡObjective:To evaluate the diagnostic value of TCM tongue in Coronary Heart Disease(CHD)patients with clopidogrel resistance after Percutaneous Coronary Intervention(PCI).Methods:To explore the independent risk factors of clopidogrel resistance in CHD patients after PCI by comparing the clinical data,laboratory examination,the distribution of lesion coronaries between clopidogrel resistance group and control group.Analysis of the characteristics of tongue color,tongue texture,sublingual vein,color and feature of tongue coating in patients with clopidogrel resistance and evaluate the accuracy of diagnosing clopidogrel resistance by TCM tongue diagnosis.Results:The history of hypertension and fibrinogen were independent risk factors of clopidogrel resistance in CHD patients after PCI.There were significant differences between the two groups in tongue color classification and sublingual venation grade.Both tongue color classification and sublingual venation grade were diagnostic parameter for clopidogrel resistance in CHD patients after PCI.The diagnostic power was increased after combining the two parameters and further improved after combining the two parameters with the history of hypertension and fibrinogen.Conclusion:TCM tongue diagnosis is helpful in the diagnosis of clopidogrel resistance in CHD patients after PCI and is wealthy of further researching. |
