Diabetic peripheral neuropathy(DPN)is one of the most common complications of diabetes mellitus(DM),which seriously impacts the life quality of diabetic patients.So far.due to the unknown pathogenesis of the DPN,both prevention and treatment of DPN remains a challenge.The traditional Chinese medicine Jinmaitong(JMT)is effective on the treatment of the diabetic patients with DPN by unknown mechanism.This study is to investigate the potential role of JMT in sciatic nerve remyelination and whether the Wnt/?-catenin pathway is involved.In this study,streptozocin(STZ)induced diabetic rats and high glucose medium cultured Schwann cells(SCs)were chosen to simulate the damage of DPN.Quantitative real-time polymerase chain reaction(RT-qPCR)and Western Blotting(WB)were used to test the mRNA and the protein expression of Wnt family member 3a(Wnt3a),Wnt inhibitory factor 1(Wif-1),glycogen synthase kinase 3?,?-catenin and myelin protein zero(PO),and the phosphoiylated protein of GSK3 ? and ?-catenin in both rat sciatic nerve and SCs.Then enzyme-linked immunosorbent assay(ELISA)was used to detect the expression of Wnt3a and Wif-1 protein in both rat serum and medium.The results showed that with the extension of time,the diabetic rats sciatic nerve injury was occurred;the Wnt/?-catenin signaling pathway was inhibited in both diabetic rat sciatic nerve and SCs in high glucose medium;the expression of' PO was decreased,and the myelin repair was inhibited.JMT was able to promote the expression of' PO in both sciatic nerve and high glucose cultured SCs by increasing the proliferation rate of SCs in high glucose medium and by activating the Wnt/?-catenin signal pathway.The increased expression of PO was able to reduce the pathological damage of those diabetic rats,and was able to promote the regeneration of the sciatic nerve myelin.10-time-human-dose JMT was the most effective dose f-or the diabetic rats and the high glucose medium cultured SCs.This study indicated that the traditional Chinese medicine JMT could ease the injury on both diabetic rat sciatic nerve and SCs in high glucose medium by improving the expression of Wnt/?-catenin signal pathway.So far.these effectiveness of the traditional Chinese medicine JMT are not yet reported in China and aboard.
