Objective: Through the experiment observation nephritis xiaobaikeli to adriamycin nephrosis rats proteinuria and Podocyte Podocin, Alpha 3 beta 1 integrin, Alpha actinin-4,the influence of nephritis xiaobaikeli were discussed from molecular biology mechanism of action.Methods: Use disposable tail intravenous injection of adriamycin glomerulonephritis proteinuria model,divided into blank control group,model control group,nephritis xiaobaikeli high-dose group, nephritis xiaobaikeli low dose group, lotensin group 5,7 weeks observation,detection of rat urine creatinine, serum albumin,urea nitrogen,serum creatinine,urine albumin immune transmission turbidimetry test,immunohistochemical detection of protein involved in the rat kidney podocytes Podocin,alpha 3 beta 1 integrin and alpha actinin - 4 average surface density, Real Time PCR detection of rat kidney Podocin mRNA,rt-pcr detection in the rat kidney 3/alpha beta 1 integrin mRNA and alpha actinin-4mRNA expression.Results: 1 .Nephritis xiaobaikeli can improve elimination adriamycin nephrosis rats general state;2.Nephritis xiaobaikeli can improve elimination adriamycin nephrosis rats kidney pathological state;3. 7 weekend each treatment group (lotensin and nephritis xiaobaikeli high and low dose group)adriamycin nephrosis rats urinary albumin/uric creatinine ratio compared with model control group significantly decreased(P<0.05),nephritis xiaobaikeli high dose group with the lotensin group compare is more obvious(P<0.05);4. 7 weekend each treatment group serum albumin levels increased significantly with the model group compare(P<0.05),nephritis xiaobaikeli high dose group with the lotensin group compare is more obvious(P<0.05);5.Immunohistochemistry showed that each treatment group in renal tissue Podocin protein expression with the model group compare of the obvious increase(P<0.05),nephritis xiaobaikeli high dose group with the lotensin group compare is more obvious(P<0.05); in the kidney tissues Alpha 3 beta 1 integrin protein expression was significantly lower with the model group compare(P<0.05),nephritis xiaobaikeli high dose group and lotensin group no significant difference(P>0.05);in the kidney tissues Alpha actinin-4 protein expression was significantly lower in the model group(P<0.05),nephritis xiaobaikeli high dose group with the lotensin group compare is more obvious(P<0.05);6.Real Time PCR showed nephritis xiaobaikeli high-dose group Podocin mRNA expression significantly raised compared with the model group(P<0.05);Rt-pcr showed that treatment group in the kidney tissues alpha/beta 1-3 integrin mRNA expression significantly raised with the model group compare(P<0.05),nephritis xiaobaikeli high dose group with the lotensin group compare is more obvious(P<0.05);in the kidney tissues Alpha actinin-4 mRNA expression significantly lower in the model group(P<0.05),nephritis xiaobaikeli high dose group with the lotensin group compare is more obvious(P<0.05);7.No effect for Renal function (P>0.05).Conclusion: 1 .Nephritis xiaobaikeli can reduce adriamycin nephrosis rats urinary protein excretion; 2.Nephritis xiaobaikeli can improve the level of adriamycin nephrosis rats serum albumin;3.Nephritis xiaobaikeli may through increase the kidney tissues of adriamycin nephrosis rats Podocin protein and Podocin mRNA expression, reduce alpha 3 beta 1 integrin protein expression in renal tissue and increase alpha 3/beta 1 integrin mRNA expression,cut in the kidney tissues of alpha actinin-4 protein and mRNA expression of alpha actinin-4 from multiple parts to repair damage to cells,maintain the integrity of the Podocyte cell structure and function,reduce urinary protein excretion.
