Background: Diabetes has become a common disease in the world,and is rapidly spreading.According to the International Diabetes Federation(IDF)latest estimates,the global diabetes population has exceeded 4.15 billion mark,that is,every 11 adults,one is diabetic patients.Taiwan's diabetes patients have 178 million people,a record high.Diabetes is a very important complication of wound healing,severe and even lead to amputation,at this stage many studies have found that when diabetes occurs,there will be a variety of molecular and cellular function damage,such as growth factors,reactive oxygen species,nitric oxide,matrix metalloproteinases,micro RNAs,endothelial progenitor cells,etc.,eventually leading to poor healing of diabetic wounds.Turmeric belongs to the ginger class herbs,is a commonly used traditional Chinese medicine,with blood circulation,blood stasis,qi,pain,through the other effects.Curcumin;English name Curcumin;is the turmeric rhizome extracted from a polyphenolic compounds,taste bitter bitter,orange-yellow.In recent years,curcumin research,found that curcumin does have a variety of biological activity,including anti-inflammatory,anti-cancer,anti-bacterial,anti-oxidation,anti-aging,anti-virus,anti-ulcer,anti-mutation,Atherosclerosis,anti-rheumatism,anti-kidney damage,eliminate free radicals,lowering blood pressure,blood pressure,hypoglycemic and so on.Lithospermum(scientific name: Lithospermum erythrorhizon)for Lithospermaceae Lithospermum plants.Lithospermum root,Ingredients: for the pigment composition,naphthoquinone derivatives,containing shikonin,elixir,acetyl shikonin,Lithospermum deoxygenation,fatty acids and Lithospermum polysaccharides and so on.Pharmacological effects: in the antibacterial aspects,for Staphylococcus aureus,Escherichia coli,Salmonella typhi,Shigella,skin fungus and influenza virus are inhibited.Oral or topical aspects of anti-inflammatory effects,acute exudation of inflammatory vascular hyperthyroidism.The preparation of ointment from the extract of Lithospermum is a topical medicinal application,which has the effect of hyperplasia of granulation tissue,and can obviously accelerate the healing ability of wound and the effect of mild antipyretic.Electrospinning(Electrospinning)in recent years has been widely used in various medical fields,such as the application of tissue engineering scaffold,skin repair materials,drug release,vascular replacement and other fields.Objective: Diabetic patients often reduce the secretion of growth factors,and its ability to reduce cell migration and migration,decreased protein secretion and other defects,so that the wound is not easy to heal,during the repair,and often due to poor care or dressing improper selection,may make the wound Bacterial infection,replacement of dressing when the secondary damage,to extend the recovery time,therefore,in order to reduce wound infection and accelerate wound repair,choose a suitable for good wound skin is necessary.In this study,the nanofiber film was prepared by electrospinning technology to simulate the extracellular matrix,promote cell attachment and growth,and add curcumin and Lithospermum to the material for the study of injured skin tissue regeneration.As the feasibility of wound dressing,and is expected to be practical in clinical applications,in order to achieve shortening the wound healing time.Methods:(1)First,different concentrations of curcumin and fibroblasts(L929 cell line)co-culture,cell survival analysis,supplemented by curcumin free machine clearance rate and tyrosine inhibition rate,to find the best The concentration of curcumin.(2)The optimum concentration of turmeric was added to the gelatin and polyvinyl alcohol solution,and the nanofiber material was prepared by electrospinning.The physical and chemical properties and biocompatibility of the materials were evaluated respectively,including the morphology of nanofibers,Structural analysis,Fourier transform infrared spectroscopy,drug release analysis,cell attachment observation,cytotoxicity test(3)Animal experiments on the implantation of diabetic mouse skin materials to further assess the in vivo biocompatibility of the wound dressing and the ability to promote tissue repair.Results:(1)curcumin concentration of less than 2?g / ml,L929 did not promote the proliferation or inhibition of the effect,but when the curcumin concentration of more than 20?g / ml,then L929 significant toxicity.(2)curcumin concentration at 2?g / ml began to have free radical scavenging effect,the concentration reached 2mg / ml the best.(3)curcumin concentration at 20 ng / ml ~ 20?g / ml with inhibition of tyrosinase effect,which the concentration of 2?g / ml when the best results.(4)SEM images of each group of electrospun film and L929 cell line for 2 days showed that the pseudopodia of L929 cells had been extended,indicating that the cells of each group had good effect and the material had biocompatibility.(5)After implantation of mouse skin for 2 weeks,the repairing effect was Gel / PVA / curcumin group> Gel / PVA group> Gel / PVA / Lithospermum group> Gel / PVA / Lithospermum / Curcumin group> Control(P <0.01),and the difference between the control group and the artificial skin group(P <0.01).(6)After 2 weeks of surgery,the content of TGF-? in the skin of mice was removed,and the gel / PVA / curcumin group was the largest and the blank group was the least.Conclusions: 1.In this experiment,two kinds of traditional Chinese medicine(turmeric and Lithospermum)in the electrospinning have obvious effect on the repair of diabetic wound.The preparation of this material is very innovative and does achieve the desired effect.2.Curcumin has anti-inflammatory,anti-cancer,anti-oxidation,anti-aging,anticoagulation,hypoglycemic and other effects,Lithospermum has antibacterial,accelerate wound healing,combined with the advantages of two traditional Chinese medicine,Meet the clinical requirements.3.Curcumin can promote the production of transforming growth factor ? and collagen protein,it can accelerate the healing of skin wounds.4.Electrospinning of the nanofiber structure can regulate the stable release of drugs 5.Gel / PVA / curcumin group> Gel / PVA group> Gel / PVA / Lithospermum group> Gel / PVA / Lithospermum / Curcumin group> Control group > Artificial skin group.6.This study can be seen to add curcumin prepared by the electrospinning film with the potential to repair the wound,but its economic value and whether it is worth mass production,remains to be assessed.
