Objective:Rg1 that is a kind of traditional chinese medicine has pharmacological effects of antiapoptotic,antifibrosis,antioxidant and estrogenic effect.is used to treat POF.Compared with MHT.It has less side effects and could restore reproductive function in the early stages of POF.POF induced D-gal in mice was established.We hope to clarify its mechism and open a new way of treatment of POF by exploring dose and method of Rg1 on POF mice modelMethed:In order to screening out optimum dose of D-galactose induced POF.C57 female mice were divided into four groups randomly,which were the control group,the low-dose group,the middle dose group,the high dose group.Body weight was recorded.Overall ovarian function including estrous cycles,sex hormone secretion,ovarian follicle development was analyzed by H&E staining and ELISA.In order to detect the therapeutic effect of Rg1,C57 female mice were divided into four groups randomly,which were the saline group,the D-gal+Rg1 group,the Rg1 group and the D-gal group,Body weight was recorded.estrous cycles,sex hormone secretion,ovarian follicle development,uterus and ovarian morphology was analyzed by H&E staining,transmission electron microscope and ELISA.Effect of Rg1 on aging was determined by analyzing the activities of oxidation-associated biomarkers,pro-inflammatory cytokine secretion,expression of senescence-associated proteins,and fertilityResults:The middle dose group reached the diagnostic criteria of POF.Compared with the D-gal group,in Rg1 +D-gal group,uterine and Ovary weight coefficient was increased significantly,estrous cycle block was released,fertility and the morphology of ovaries and Uteruses were restored.Rg1 treatment after D-gal administration significantly reduced senescence-associated protein expression,increased the activity of total superoxide dismutase and glutathione peroxidase from bovine erythrocyte,and induced higher follicle stimulating hormone receptor protein expression.Additionally,the expression levels of malondialdehyde,interleukin-1b,tumor necrosis factor-a,and interleukin-6 were significantly decreasedConclusion:Rg1 improves mice fertility and reduces ovarian and uterine pathological damage in D-gal-induced POF model possibly through enhancing anti-inflammatory,anti-oxidant capacities and increasing expression of FSHr proteins in the granular cell and reducing expression of senescence signal pathway proteins.
