Among avian metabolic disorders,Tibial dyschondroplasia?TD?has wide prevalence worldwide in birds having fast growth potential especially in broiler chickens.It is characterized by non-mineralization and avascularization of growth plate?GP?,and led to lameness and uncharacteristic growth of chondrocytes.Recently reported that TD account30%of all the bone diseases in poultry and morbidity rate was approximately 10%in China which caused a serious reduction in the farmer's profit margin.Clinical and subclinical effects of TD include immunity reduction,osteomyelitis,less growth rate and breast cyst formation,that in terms upraise the animal ethical and welfare issues and degrade the chickens'carcass and meat quality.1. Recovery of Chickens Affected with Tibial Dyschondroplasia by Application of Grape Seed Extract Through Downregulating CA2 Gene and Enhancing Liver FunctionsGlobally Tibial Dyschondroplasia?TD?is a serious metabolic syndrome in broilers that is identified by the accretion of the growth plate?GP?into the tibial metaphyseal portion.Grape seed extract?GSE?is a well-known significant antioxidant since it possesses multifaceted anti-inflammatory,anti-cancer and reducing reactive oxygen ability.The experimental test was intended to evaluate the proficiency of GSE to get well the chicks affected with TD.For this purpose,210 chicks were bought and fed normal starter feed for the first 3 days.After that two groups were made having 70?Control;normal feed?and 140 chickens?Thiram;normal feed+thiram,50 mg/kg?.Further two equal divisions were made in Thiram group on 7th day viz TD group and GSE group?50 mg/kg/day?.GSE significantly?P<0.05?reverted the blood serum profile and liver antioxidants metabolites and,reduced the histopathological abrasion of the liver as compared to the TD group.Furthermore,GSE chicks exhibited a significant decrease?P<0.05?in CA2gene expression and less mortality,TD score and GP size.In inference,GSE is a natural plant-based extract which owns solid sifting,bone healing property and strong antioxidant capacity.GSE provided recovery to chicks affected with TD chicks via dropping TD score,whereas stabilized the GP width through the CA2 m RNA and protein downregulation.2. Effect of Grape Seed Extract on Tibial Dyschondroplasia Incidence,Liver Weight,and Tibial Angiogenesis in ChickensAmong avian metabolic disorders,Tibial dyschondroplasia?TD?has a wide prevalence worldwide in chickens that are identified by non-mineralization and avascularization of the growth plate?GP?.Grape seed extract?GSE?reduces the inflammation and oxidative stress by scavenging free oxygen specie and has some anti-cancer and angiogenic activity.This study was done to evaluate the role of GSE for the therapy of the chickens affected with TD.Arbor Acres broiler chicks?n=210?weighing 47±6g were fed starter feed till first 3 days and then portioned equally into three groups?Control,TD and GSE group;n=70 each?.Thiram was incorporated in the feed of TD and GSE chicks until day 6 for inducing tibial dyschondroplasia?50 mg/kg of feed?,whereas usual feed was offered to control group.All the birds in TD and GSE group showed the symptoms of TD on day 7and GSE group was given Grape seed extract?50 mg/kg/day?intraperitoneally till the termination of the experiment.Chickens fed with thiram-feed exhibited high TD score and GP measurement.After the GSE treatment,the TD score was dropped remarkably and,inflammation and TD lesion was reduced.Moreover,body weight gain and liver weight were increased in comparison with TD chicks after application of GSE.GSE supplementation alleviated the adverse effects of thiram and increase in angiogenesis was observed.In conclusion,GSE recovered the TD affected broilers through downregulating the HIF-1?and Hsp90 expressions.3. Plastrum Testudinis Extract Mitigates Thiram Toxicity in Broilers via Regulating PI3K/AKT SignalingTibial dyschondroplasia?TD?negatively affect broilers over the world,in which accretion of the growth plate?GP?take place into tibial proximal metaphysis.Plastrum testudinis extract?PTE?is famed as a powerful antioxidant,anti-inflammatory,and anti-cancer agent.This trial was conducted to assess the efficiency of PTE to recover the TD affected chickens.300 chicks?day old?chicks were raised for 3 days with normal feed.On 4th day three groups?n=100 each?were made namely Control?normal diet?,TD and PTE group?normal diet+thiram,50 mg/kg?.On 7th-day thiram was stopped in TD and PTE group,and PTE group received normal diet and PTE?30 mg/kg/day?.PTE revert the liver antioxidants enzymes and serum biochemicals in TD chickens.Furthermore,PTE lessened the mortality,TD score,TD lesions and GP size,and increased the AKT and PI3K genes expression in TD chickens.In conclusion,PTE has the potential to heal the bone and act as a strong antioxidant agent.PTE can recover the TD affected broilers by decreasing TD score and regulating GP size via upregulation of the AKT and PI3K m RNA and protein expressions.
