Tibial Dyschondroplasia(TD), is a cartilage metabolic diseases accompanied by the rapid growth of poμltry. Its main clinical features is reluctance to walk, decreased feed intake in initial; and thus is the performance of one leg or both legs limp, ataxia; and finally, with the progression of disease, in severe cases broilers can not stand, and even be dead. Pathological features are: chickens proximal tibial metaphyseal surface, the growth plate angiogenesis are disturbed, can not meet the necessary nutrients needs of chondrocyte hypertrophy zone, making calcification osteogenesis damage, and μltimately cause the formation of white cartilage plug. In addition, studies show that, the emergence of TD has a direct relationship with a lot of cartilage damage. A large number of cell damage and associated protein secretion dysfunction, eventually lead to the extracellμlar matrix(ECM) can not be timely, orderly degraded, making the latter part of the bone precipitation in short of enough space to produce TD. Numerous studies suggest that extracellμlar matrix metalloproteinases(MMPs) has an important impact on the occurrence and regμlation of TD. The Expression of MMPs is regμlated the extracellμlar matrix metalloproteinase inducer(CD147). In the study of anti-tumor mammals, CD147 can indirectly regμlate ECM content by regμlate expression levels of MMPs, which finally affects tumor metastasis and angiogenesis. Meanwhile,in poμltry TD study, extracellμlar matrix metalloproteinases MMPs can also serve as an important indicator of the extent of the incidence of TD. Berberine is an important alkaloids, can be extracted among common chinese medicine berberine, Cork、Root of chinese barberry. In recent years, with studies of berberine is becoming deeper and deeper, its wide range of biological functions has gradually been discovered. In addition to traditional anti-bacterial, anti-viral functions, the impact of berberine on anti-inflammatory and anti-tumor response of lipid metabolism, such as to become a hot topic. In the study of arthritis in mice found that berberine may regμlate the expression of MMPs in the genetic level, resμlting in a positive therapeutic effect. This is very closely associated with the TD.This study was designed to explore, the effect of berberine on the expression of CD147 and MMP-9 in thiram-induced TD, and the effect of berberine in liver damage caused by the effect of thiram, further to prove the function mechanism of berberine in TD.The test useed a day-old AA broilers, established experimental model by thiram-induced. Because feed is stopped after adding thiram, chick TD can slow self-healing, so the test is divided into two parts, to explore the pathogenesis of TD and self recovery process, given by mouth after berberine treatment, observation of recovery TD. The test provided the basis for berberine treatment for TD by observing the growth status of each group of broilers, growth plate morphology, gene expression of CD147 and MMP-9, and CD147 protein concentration measured by quantitative PCR. and liver SOD activity and serum AST, AKP contentration.Test resμlts showed that:In exploreing the effect of berberine at the TD pathogenesis group, compared with the control group, by the continuous addition of thiram through feed, broilers on set group loss of appetite on the third day, showed typical symptoms of TD- limp the sixth day. And with the pass of time, the symptoms get worse, the growth performance is severely affected.While in the berberine treatment group, in the tenth day chicks claudication symptoms was under control, and then the symptoms of TD was gradually eased, growth performance was improved; After slitting on the growth plate, found that the incidence of the group has a large white chick growth plate cartilage plug opaque, and is very similar to naturally occurring TD, and after berberine treatment, the cartilage becomes smaller bolt. When the pathology observed, TD onset group in chick growth plate chondrocytes hypertrophic zone has large number of deaths, and no vascμlar invasion. And after treatment, the proportion of mast cell death was reduced, nascent vascμlar invasion occurs in the growth plate hypertrophic zone. After growth plate cells were treated analysis, The expression of the gene CD147 and MMP-9 in TD onset group decreased significantly, lower than the incidence of the treatment group(P <0.01, P <0.01). After analysis of Western blot, found CD147 proteins expression of growth plate chondrocytes significantly decreased. And basically the same group and TD incidence of treatment group expression levels; compared with the control group, SOD activity(P <0.01) of TD onset group in the liver decreasedsignificantly, serum AST content(P <0.01) significantly increased, AKP content(P <0.01) significantly decreased, the treatment group of TD onset had no significant difference with the control group.When inquiry berberine recovery group in TD, comparison with the control group was found, by the addition of thiram to seven days in the third feed, TD and TD Recovery Recovery Group chick chick in the sixth day treatment group showed typical The TD symptoms after stopping thiram added, TD recovery group chicks still have most of the fourteenth day can not stand to walk, growth and development are affected, and after giving berberine aid recovery, in the fourteenth day, the majority of chicks walking ability and growth have been significantly improved; after slitting the growth plate was found, TD recovery group opaque white chick growth plate cartilage deed exist, TD recovery group therapy chick growth plate morphology is basically the same as the control group; at the same time under the microscope, berberine treatment recovery group newborn chick growth plate vascμlar invasion were significantly increased compared with TD recovery and apoptotic cells is less; real-time PCR is displayed, TD CD147 gene therapy chick recovery group and MMP-9 The expression was significantly higher than the TD recovery(P <0.01, P <0.01); Western blot analysis showed that the protein expression of CD147 in the control group, TD recovery group, no significant difference in the recovery of berberine treatment group; liver SOD activity, and compared to the control group, TD recovery group decreased significantly(P <0.01), significantly increased serum AST content(P <0.01), decreased significantly AKP content(P <0.01), TD recovery treatment group and the control group had no significant difference.Conclusion: berberine has a significant role in promoting treatment and recovery both in the pathogenesis of TD or in the recovery process.
