Salmonella is an important zoonotic pathogen,there have been a variety of drug-resistant strains in the world,a direct threat to human public and food safety.Salmonella mainly infects humans and animals in the form of foodborne transmission,causing some related gastrointestinal diseases,and in serious cases can lead to systemic infection.The use of antibiotics induces the emergence of bacterial drug resistance.In the livestock and poultry industry,pullorosis continuously harms the development of chicken industry and causes serious economic losses to poultry in our country.Antibiotics are the main method to treat salmonella infection,but the widespread use and abuse of antibiotics lead to the emergence of bacterial drug resistance and drug residues.Therefore,there is an urgent need for a new anti-infection strategy to solve the increasingly serious problem of drug resistance.Salmonella mainly has two sets of virulence systems SPI-1 and SPI-2.Virulence island 1 plays an important role in the process of salmonella invading host cells.Virulence Island 2 plays an important role in the host cell colonization of Salmonella Type III secretory system(T3SS)exists conservatively in Gram-negative bacteria and plays a key role in the process of bacteria infecting the host.Because T3 SS is not a necessary gene for bacterial growth,by inhibiting the function of T3 SS,bacteria are removed from the body through the immune defense system without affecting bacterial growth.Therefore,in the process of salmonella infection,taking T3 SS as the drug target can achieve the purpose of anti-salmonella infection.Under the background of food safety,medicine safety,the development of Chinese herbal medicine for the prevention and treatment of infectious diseases with has attracted much attention.Our research group has been engaged in the screening and mechanism of inhibitors of traditional Chinese medicine and its active components targeting important bacterial virulence factors for a long time,which provides a new strategy and potential lead compounds for traditional Chinese medicine.A series of inhibitors were screened in natural compounds,and it was found that cinnamaldehyde could significantly reduce the pathogenicity of Salmonella in vitro and in vivo by interfering the function of bacterial type III secretory system without affecting the growth of Salmonella.Cinnamon oil is a volatile oil obtained by steam distillation of dried branches.It is a natural medicine extract.The main component of cinnamon oil is cinnamaldehyde,the content is more than 85%.Therefore,this project uses cinnamon oil as raw material,through systematic prescription screening,preparation process investigation,establishment quality detection method,sample stability study,etc.Successful obtain cinnamon oil oral liquid,provide guarantee clinical research data for pharmacology,toxicology and expand clinical research.In this study,through the establishment of salmonella type ? secretion system report system,through detection the type ? secretion system activity,it was found that cinnamon oil oral liquid could inhibit its function,and had no inhibition of bacterial growth and bactericidal activity.According to the evaluation of drug activity in vivo and in vitro,it was found that cinnamon oil oral liquid could significantly reduce the ability of Salmonella typhimurium and Salmonella pullorum entry into host cells,To further clarify the mechanism of inhibitory effect of cinnamon oil oral liquid on type ? secretion system of Salmonella typhimurium.In this study,biochemical tests in vitro and molecular biology methods were used to verify that the yield of SipA,SipB and regulatory protein HilA in type ? secretion system of Salmonella typhimurium treated with cinnamon oil oral liquid decreased significantly.RT-PCR experiment confirmed that cinnamon oil oral liquid affected the upstream regulation of Salmonella type III secretory system and further affected the expression of downstream proteins.Artificial infection and enlarge clinical infection of salmonella in chickens,it was found that cinnamon oil oral liquid was the best dose for the treatment of salmonellosis in chickens was 0.18g/kg body weight / day,and the preventive effect was better.General clinical condition,detection of blood physiological index and serum biochemical index were detective in safety research and evaluation.The safety of broilers was studied and evaluated from the aspects of general clinical condition,blood physiological index detection,serum biochemical index detection and so on.The results show that cinnamon oil oral liquid is safe at the recommended dose of 1,3 and 5 times for 10 days,and it can be used in clinic according to the actual situation.To sum up,cinnamon oil oral liquid can inhibit the activity of type ? secretory system by affecting the transcriptional regulation of type ? secretory system of Salmonella without affecting the growth of bacteria.Through the evaluation of drug activity in vivo through the animal model of artificial infection,it was found that cinnamon oil oral liquid had a protective effect on the death of chicken caused by Salmonella pullorum.In this study,molecular genetics,biochemistry and other experimental methods were used to verify the mechanism of cinnamon oil oral liquid against salmonella type ? secretion system,and to provide new therapeutic drugs for the application of innovative drugs for Salmonella infection in livestock and poultry in China.
