Objective:To explore the change of gene expression profilek between anti cardiolipin antibody positive patients with inevitable abortion,and explore the conduction mechanism of TLR4/nf-kappa B signaling pathway.To reveal the effect of traditional Chinese medicine and cryptotanshinone against cardiolipin antibody lead to recurrent abortion.And reveal the relationship between TLR4/nf-kappa B signaling pathway and recurrent abortion.Methods:1.The article uses gene chip technology to detect the gene expression of ACA(+)recurrent miscarriage which is profilekidney and blood stasis type and normal early pregnant women,and screening different expressed genes.And to analyze their information,confirming the conduction mechanism of TLR4/nf-kappa B signaling pathways.2.According to the serum pharmacology method extracted from rats serum of the drug-containing serum,to establish the system of decidual stromal cells in vitro.And then give them drug-containing serum and cryptotanshinone respectively to observe changes of TLR4/nf-kappa B signaling pathways.At last to reveal the regulating mechanism of traditional Chinese medicine and cryptotanshinone at TLR4/nf-kappa B signaling pathways.Results:1.Gene chip can screen different genes between ACA(+)recurrent miscarriage which is kidney and blood stasis type and normal early pregnancy,and the number of genes is 851.Compared with the control group 234 genes increase,and 617 genes cut.2.Two groups have many differences such as cytokine-cytokine receptor interaction,glutamatergic synapse,osteoclast differentiation,protein digestion and absorption,mineral absorption,retinol metabolism,cytochrome p450 of xenobiotics metabolism,the cytochrome p450 drug metabolism,et al.3.The number of different signal pathways is 5,for example TLR4/nf-kappa B signaling pathways,calcium signaling pathways,PI3K-Akt signaling pathway and MAPK signal pathway,p53 signaling pathway.4.TLR4/nf-kappa B signaling pathways involves in the ACA(+)recurrent miscarriage which is kidney and blood stasis type.5.To analyse TLR4/nf-kappa B signaling pathways of ACA(+)recurrent miscarriage which is Kidney deficiency and blood stasis type.There are three genes increased,such as TLR4,the nf-kappa B and TNF-a.There are one gene cut,and it is COX2.6.Tonifying kidney and promoting blood circulation decoction and Implicit tanshinone can cure recurrent miscarriage and inhibite TLR4/nf-kappa B signaling pathway.7.The treatment efficacy of Tonifying kidney and promoting blood circulation decoction is best,and The second is cryptotanshinone.Conclusion:1.Gene chip can screen different genes between ACA(+)recurrent miscarriage which is kidney and blood stasis type and normal early pregnancy 2.According to TLR4/nf-kappa B signaling pathways the anti cardiolipin antibody can lead to recurrent miscarriage.3.Tonifying kidney and promoting blood circulation decoction,cryptotanshinone and aspirin can treat ACA(+)recurrent miscarriage early inevitable abortion,but the treatment effect of Tonifying kidney and promoting blood circulation decoction is the best.
