Chinese medicine theory of nature and flaour is an important part of traditional Chinese medicine,highlights the characteristics of Chinese medicine theory and also highly summarizes clinical diagnosis and treatment for prescription drug removal experience.Nature and flaour theory is also a concentrated expression of human interaction with the drug and summary results,the content of which is rich and complex.Stephania tetrandra S.Moore is a TCM.And the record of clinical application has more than 2000 years.In long application process,on one hand,the content of nature and flaour theory of Stephania tetrandra constantly enriched and developed;on the other hand,there are some inevitable mistakes in literature recording and the efficiency heritage.Among the differences,modem scholars have some questions on the realization of nature and flaour which also affect guiding role in nature and flaour theory of Stephania tetrandra in treatment.Based on this,creatively using the nature and flaour theory of traditional Chinese medicine hypotheses provided by Professor Haixue Kuang.With the corresponding"Chinese nature and flaour can be split and combined" mode is also provided,which is the first time to research on nature and flaour of Stephania tetrandra.This is not only exploring the content of nature and flaour of Stephania tetrandra,but also verifying scientific connation of nature and flaour of Stephania tetrandra in different levels and aspects,which also develop the realization of theory and effectiveness of Stephania tetrandra and enrich related knowledge to Chinese medicine.1.Through analyzing and tidying the representative ancient herbal medicine book(time span in 2200),we make an study on the nature and flaour of Stephania tetrandra and find that first appearance of Stephania tetrandra was "piquancy and flat".And at this stage,the nature and flaour is "bitter and cold".Carding the evolution progress of Stephania tetrandra in system,confirm the awareness of Stephania tetrandra in different periods and analysis of the connotation and formation of the root causes of cognitive differences,illustrate the correlation between changes in taste and clinical efficacy and application.2.Establish a progress that full composition do not mutually cross the split process.Using boiling water reflux extraction method to get water decoction and comprehensive using differential solubility,liquid-liquid extraction and ion exchange resin technology,split components were divided into alkaloid component,non-alkaloid component,polysaccharide and steroid component.By HPLC,UPLC-MS and GC-MS methods,characterize the chemical composition,to clarify its material basis,by PCA and OPLS-DA multivariate mathematical statistics,which do not cross validation.Meanwhile,it is the first time to use RSM method to optimize polysaccharide extraction methods by Box·Behnken design to get optimal extraction process.3.From six pharmacodynamics,establish pharmacological evaluation index of Stephania tetrandra by twelve set of experiments,which make the "split component of nature and flaour ","fiaour" and "concrete effect" associated.Using anti-inflammatory(swelling suppress acute inflammation,reduces cytokine release),analgesia(peripheral inhibition,CNS depression),antipyretic(anti-pyrogenic elevated body temperature),diuretic(increase urine discharge),immunomodulators(unspecialized heterosexual immunosuppression,specific immunity humoral immune regulation,immune suppression of specific immune cells)and anti-rheumatic(rheumatoid arthritis,gouty arthritis)evaluate the flaour which have been assigned by TCM combined Clustering Analysis.It is clear that the flaour of Stephania tetrandra are bitter and pungent.The features of bitter reflect anti-inflammatory,analgesic,antipyretic,diuretic,immunosuppression and anti-rheumatic,and the material basis are alkaloids component.The features of pungent reflect analgesic,antipyretic,diuretic and immunosuppression,and the material basis are sterides,non-alkaloid and polysaccharide split components.4.Through metabolism,energy metabolism and central nervous system neurotransmitter,the "split component","property "and" metabolism" are associated.Evaluate the medicinal properties on metabolism T3,T4,TSH,TRH,17-OHCS,cAMP,cGMP,PA,PDH,IUPAC,LDH,SDH,Na+-K+-ATP and central nervous system neurotransmitters 5-HT,DA,NE and AchE,which were assigned by promoting and inhibiting the body’s metabolic state.It is clear that the nature is cold and flat,the function of cold is to inhibit the metabolism and reflect the energy metabolism,alkaloid component is the material basis,while non-alkaloid,steroidal and polysaccharide component are material basis of flat.5.In this paper,we use Tetrandrine which is most significant component in "bitter and coldr" as research object.As bitter has the effect of specific selection anti-tumor effect,using "hot and cold" mechanism choice cell energy metabolism,to explore pro-apoptotic activity of Tetrandrine in tumor cells and the underlying mechanisms.We selected HepG-2,BGC-823,MCF-7,A253 and SKOV-3 cells to test cytotoxic activity of Tetrandrine;and choose HepG-2,BGC-823 and MCF-7 which have significant cytotoxic activity for cell energy metabolism analysis by Seahorse XF24 Extracellular Analyzer.We found that tetrandrine has different affect on the tumor cells on the basis of oxygen consumption,mitochondrial capacity,mitochondrial energy storage and proton leak in four areas.Further illustrated,we use the mitochondrial membrane potential,adenosine triphosphate,the active oxygen content and respiratory electron transport chain complex Ⅰ-Ⅳ enzyme vitality(NADH dehydrogenase,succinate dehydrogenase,eytochrome reductase and cytochrome oxidase),which may be found to affect mitochondrial function.It is clear that antitumor-mechanism of tetrandrine is destroying the mitochondria,reducing cellular energy metabolism,which play a role in promoting tumor cell apoptosis,meanwhile nature and flaour of Tetrandrine is bitter and coldIn summary,new hypotheses of nature and flaour has been combined with the corresponding research model,property and taste of Stephania tetrandra to verify and practices.Establish the split progress of Stephania tetrandra to fulfill full split component do not cross between taste components and keep the initial chemical composition and structure.Establish nature and flaour pharmacology evaluation system and evaluate and assign the taste and property of Stephania tetrandra At the same time,the theory of taste has been extended to the monomer compounds,and guide the anti-tumor effect and mechanism.The ideas and methods are standardized and scientific,and we can provide a reference for future studies of the same type.
