Objective:This paper is based on Professor Kuang Hai's "one flavor one property,and X flavors Y properties(Y<X)" new hypothesis on traditional Chinese medicine flavor and property theory.On basis of decoction from Stephania tetrandra on rheumatoid arthritis in Bovine ? collagen-induced arthritis model and Uric acid sodium salt crystal induced gouty arthritis in rats,we attempted to clarify substance basis of "bitter" taste effective from the traditional Chinese medicine fen fangchi(S.7),and evaluated its taste pharmacology study and further deepen our understanding of the nature of smell,descripe the relationship on smell and arthritis.Methods:1.Study on the therapeutic effect on decoction of rheumatoid arthritis in Bovine ?collagen-induced arthritis model in rats.By foot,tail,back intradermal injection of bovine type ? collagen emulsion.7 days later to strengthen the injection,establishment of CIA rat model.Design the control group,model group,tripterygium glycoside(0.0015 g · kg-1),indomethacin group(0.0135 g · kg-1)and decoction of low,middle and high dose group(0.6831 g · kg-1,1.3662 g · kg-1,2.0493 g · kg-1).Respectively filling stomach administration,continuous administration 21d,once per D.Study on the arthritis index,toe swelling in rats,IL-1,IL-1? IL-6,TNF-a levels in serum and joint pathological changes in other indicators.2.Study on the Uric acid sodium salt crystal induced gouty arthritis in rats,design the control group,model group,colchicine(0.0015 g · kg-1)and decoction of low,middle and high dose group(0.6831 g·kg-1,1.3662g·kg-1,2.0493 g·kg-1).Respectively filling stomach administration,continuous administration 21d,once per D.The 7d gavage after crystallization of sodium urate suspension into the right ankle joint cavity made of gout rat model.In this paper,we research that whether decoction had arthritis treatment effect by their arthritis index,toe swelling,serum lipids in rats quality peroxidation(SOD,MDA,GSH-PX),rat kidney,liver and spleen indices,pathological changes in the joints of rats and other indicators.On this basis,immunohistochemical method was used to detect the expression of synovial tissue of rats TLR4,MyD88,NF-KBp65 in decoction of gouty arthritis in rats.Results:1.In the experiment of rheumatoid arthritis(RA),To compare with control,model group had highly significantly levels in rats arthritis index,swelling rate and IL-1,IL-1?,IL 6 and TNF-a in serum(P<0.01).There was a varying degrees of reduction(compared with model group)in decoction low,middle and high dose group onrats arthritis index,swelling rate and IL-1,IL-1?,IL 6 and TNF-a in serum.decoction high dose group had significant difference(P<0.01).Treatment effect of joint tissue biopsy can be visually observed,which can effectively control the synovial endothelial cell damage,inhibition of pannus proliferation.2.1n the experiment of gout arthritis(GA),To compare with control,model group had highly significantly levels in rats arthritis index,swelling rate and expression of TLR4?MyD88?NF-KBp65(P<0.01),had lower significantly levels in SOD and GSH-PX in serum(P<0.01),had a rise levels in MDA and kidney index(P<0.05).There was a varying degrees of reduction(compared with model group)in decoction low,middle and high dose group onrats arthritis index,swelling rate and expression of TLR4?MyD88?NF-?Bp65.decoction high dose group had significant difference(P<0.01).Decoction low,middle and high dose group can increase the level of SOD and GSH-PX in serum,reduce the level of MDA in serum.High,middle and low dose group can effectively control synovial endothelial cell damage and inhibit the infiltration of inflammatory cytokines,while inhibition of TLR4 is involved in recognizing pathogen evolutionary conserved antigen molecules conserved in evolution,inhibiting MyD88-dependent signaling pathway,and inhibition-related gene transcriptional activation of inflammatory cytokines,blocking a complex signaling cascade reaction,blocking the start of nuclear transcription factor NF-?B to inhibit inflammation and gout regulate autoimmunity role.Conclusion:Decoction from Stephania tetrandra of low,middle and high dose groups were able to varying degrees of relieve the clinical symptoms of rheumatoid arthritis arthritis and gouty arthritis."Bitter" taste of Stephania tetrandra S.Moore had been interpreted from the treatment of arthritis.The research deeply showed the contact of decoction and "bitter" taste of Stephania tetrandra S.Moore,which lay a solid foundation and theoretical basis for the next step of taste research on Stephania tetrandra S.Moore.
