| Objective:This topic based on the traditional Chinese medicine understanding of hypertension,through acupuncture SHR Taixi.Contrasting non-acupoint group,model group and normal WKY group,try to discover acupuncture point of onset of antihypertensive effect.And then use PET-CT imaging technology to find the target area of blood pressure lowering by detecting glucose metabolism in brain area.To explore the effect of acupuncturing Taixi point on the target area of hypotensive brain in SHR and its possible central hypotensive mechanism.To explore the time point of acupuncture,antihypertensive effect and a possible new target of central antihypertensive.To provide scientific basis for the treatment of hypertension by acupuncture and moxibustion,and also the new ideas for exploring the material way of acupuncture and moxibustion treatment.Methods:(1)Determine the regularity of antihypertensive effect of meridian points24 SPF 10-week-old SHRs(male,weighing 180-200 g,systolic blood pressure≥140mm Hg)were randomly divided into taixi group,non-acupoint group and control group with 8 rats in each group,with normal blood pressure WKY rats8 only.Taixi group and non-acupoint group rats were acupunctured respectively,the program was as follows:in the awake state of the rat fixed,acupuncture left and right acupoints,with rotation frequency of 80 ± 5 times / min,the degree of rotation 180 ± 5 degrees.Each point operated 2.5 minutes,two points a total of 5 minutes.Acupuncture was operated once a day,continuous acupuncture 7 times in all.The rats in the control group were fixed for 5minutes but indeed were not acupunctured.According to "Acupuncture and Moxibustion," the method of acupoint selection for animals,to take taixi,non-acupuncture points for acupuncture.The blood pressure was measured by using tail artery pressure method to measure the blood pressure of rats.Each rat was measured continuously for 15 times and the average blood pressure was taken.Measurement time points included blood pressure before acupuncture,30,60,90 min after acupuncture.After determining the decompression effect,the PET-CT scans were performed.(2)To confirm that acupuncture activates specific brain regions in the brain After determining the antihypertensive effect,PET-CT scans were performed on each group of rats.According to the time point of the acupuncture effect,an immediate PET-CT scan and a post-treatment PET-CT scan were performed.Immediate PET-CT scan:to determine the PET-CT scan time immediately after acupuncture based on the onset time of decompression.First fix the rat,taixi acupoint group and non-acupoint group SHR were acupunctured intervention for5 min,2.5min for each acupuncture point.Thirty minutes later,intraperitoneal injection of pentobarbital sodium and maintenance of anesthesia were performed and then brain PET-CT scans were performed.The time of tail vein injection of the developer was determined according to the acupuncture and antihypertensive onset time,and 18F-FDG developer was injected from the tail vein.Rats in the normal group and the model group were only grasped and were not subjected to acupuncture.The remaining operations were the same as those of the acupuncture group.PET-CT scan after treatment:After the completion of the acupuncture treatment(7days),PET-CT scans were performed on all groups of rats.Acupuncture intervention was not applied during the whole process of injection of imaging agent and PET-CT scan.Image acquisition and PET image analysis by using SPM8 were performed by the Institute of High Energy Physics,Chinese Academy of Sciences.Results:1.each group of rats’ weight and raw blood pressure:(1).Before acupuncture treatment,the blood pressure in model group,acupuncture at taixi acupoint group and acupuncture at non-acupoint group were higher than it in normal WKY group rats,p <0.05 which infers the difference was obvious statistically implication.The blood pressure was higher than that of normal WKY rats and SHR,which meaned the blood pressure of experimental animal models was comparable to that of normal rats.There were no significant differences in the values of blood pressure between the modelgroup,taixi group and non-acupoint group(p> 0.05).Prompt acupuncture treatment of all animal models before the value of blood pressure was not large,to ensure the the good homogeneous contrast between the experimental groups.(2).Immediate acupuncture,to measure the blood pressure 10 min after acupuncture,to observe the time-effectiveness of acupuncture antihypertensive.As shown in Figures 3 and 4,compared with normal WKY rats,the blood pressure of rats in model group was higher at 30,60 and 90 min,and there was significant difference between the two groups in blood pressure(p<0.05).WKY rats,SHR,model rats had higher blood pressure,animal model of blood pressure and normal rat blood pressure were comparable.After acupuncture at taixi point,systolic blood pressure and diastolic blood pressure of rats were lower than those of model rats at 30 and 60 min,p<0.05,which refered to the statistically obvious difference implication.The blood pressure rised over time.Prompted immediately after acupuncture taixi point,systolic and diastolic blood pressure in rats were reduced.To reduce the therapeutic effect,30 min after acupuncture at taixi buck optimal effect.After acupuncture at non-acupoints,they had no significant changes in blood pressure at 30,60 and90min(P>0.05).Prompted acupuncture non-acupoint group after 1 time,it did not achieve the effect of reducing their own blood pressure in rats.The results were as follows: compared with acupuncture at non-acupoints,the blood pressure of acupuncture at taixi decreased at 30 and 60 min,the difference was statistically significant(p<0.05).Acupuncture taixi points after 1 time,antihypertensive effect was better than the non-acupoint rats.Compared with model group,non-acupoint group had no significant changes in the blood pressure of rats,p>0.05 which meaned the difference wasn’t obvious statistically implication.Prompted acupuncture non-acupoint did not reduce the therapeutic effect of reducing blood pressure in rats.(3).After acupuncture treatment,24 h after acupuncture began to measure blood pressure,continuous measurement of 3days.To observe the timeliness of blood pressure after acupuncture,as shown in Fig.5 and 6,the blood pressure of rats in model group was higher at 24,48 and 72 h after treatment compared with that of normal WKY rats,and there was significant difference between the two groups in blood pressure(p<0.05).Compared with normal WKY rats,the blood pressure of model rats after treatment was higher,and the blood pressure of experimental animal model was comparable with that of normal rats.After acupuncture treatment of taixi acupoint(7 times),the systolic and diastolic blood pressure of rats in acupuncture at taixi acupoint was lower than that of model rats 24 and 48 h after treatment(P<0.05).The blood pressure was lowest at 24 h compared with other time points,and then blood pressure increased with time.Prompted acupuncture taixi treatment,systolic and diastolic blood pressure in rats reduced the blood pressure of rats to reduce the therapeutic effect,and acupuncture at 24 h got the best antihypertensive effect.The acupuncture at non-acupoints,blood pressure at 24,48,72 h compared with the model group,had no significant change in blood pressure,no statistically significant(p>0.05).Prompted acupuncture non-acupoint group after treatment,it did not achieve the effect of reducing their own blood pressure in rats.The results were as follows:compared with acupuncture at non-acupoints,the blood pressure of acupuncture at taixi group decreased at 24 and 48 h,the difference was statistically significant(p<0.05).Prompted acupuncture taixi group,the antihypertensive effect was better than acupuncture non-acupoint rats.Compared with model group,acupuncture at non-acupoint group had no significant changes in blood pressure,and the difference was not statistically significant(p>0.05).Prompted acupuncture non-acupoint did not reduce the therapeutic effect of reducing blood pressure in rats.2.Brain area of rats with SHR model and WKY control group had higher glucose metabolism than that of WKY control group.On the first day of treatment,rats in SHR model group had more glucose metabolism than rats in WKY control group.It showed that these brain regions were activated.Brain areas with decreased glucose metabolism were mainly concentrated in visual cortex,sensory cortex,and motor cortex while the increased glucose metabolism in the brain areas mainly concentrated in:the lower edge of the cortex,thalamus,ventral orbital cortex,dorsal thalamic lateral nucleus,dorsal thalamus,orbital cortex,hypothalamus.On the 8th day of treatment,compared with the WKY control rats,the glucose metabolism in some brain regions of SHR model rats changed.It showed that these brain regions were activated.Brain areas with decreased glucose metabolism were mainly found in caudate nucleus,basal ganglia,corpus callosum,cingulate gyrus,cingulate cortex,motor cortex,sensory cortex,visual cortex,while the increased glucose metabolism in the brain areas mainly focus on:anterior cerebellar,lower cortex,thalamic,ventral orbital cortex,dorsal thalamic lateral nucleus,dorsal thalamus,orbital cortex.3.Acupuncture between taixi group rats and SHR model group rats brain regions with changed in glucose metabolism: on the first day of treatment,rats in the taixi group had a function of activation of some brain regions compared with rats in the SHR model group.The amount of glucose metabolism changed.The brain regions with reduced glucose metabolism were mainly concentrated in pre-olfactory nucleus,nucleus accumbens,hypothalamus,thalamus,prefrontal cortex,olfactory bulb,anterior commissure,dorsal thalamus-lateral nuclei,dorsal thalamus,hypothalamus,septal area,peripheral nucleus,orbital cortex,striatum.The brain regions with increased glucose metabolism were mainly concentrated in the posterior cerebellum and visual cortex.On the 8th day of treatment,compared with rats in the SHR model group,the function of some brain regions was activated and the amount of glucose metabolism was changed in the acupunctured taixi group.The brain regions with reduced glucose metabolism mainly included:bed nucleus,thalamus,inner capsule,peripheral septal nucleus,and striatum.The brain regions with increased glucose metabolism were mainly concentrated in the lower leg and the olfactory cortex.4.Acupunctured taixi group rats and acupuncture non-acupoint group rats to compare glucose metabolism changes in the brain area:on the first day of treatment,acupuncture taixi group rats compared with acupuncture non-acupoint rats,some brain function When activated,its glucose metabolism changed.The brain regions with reduced glucose metabolism were mainly concentrated on:no granular island cortex,insular cortex,medulla oblongata,and pons bridge.The brain regions with increased glucose metabolism were mainly concentrated in the lower leg,hippocampus and hippocampus.On the 8th day of treatment,the acupuncture of rats in taixi group was compared with acupuncture of non-acupoint rats.The function of some brain regions was activated,and the glucose metabolism was changed.The brain regions with reduced glucose metabolism were mainly concentrated in the thalamus,medulla oblongata,and pons bridge.The brain regions with increased glucose metabolism were mainly concentrated in the pre-olfactory nucleus,basal ganglia,caudate nucleus,olfactory bulb,prefrontal cortex,corpus callosum,posterior cerebellum,orbital cortex,piriform cortex,and striatum.Conclusion:1.Acupuncture at taixi point has a definite antihypertensive effect on SHR,while the non-acupoints have no antihypertensive effect.Immediately after acupuncturing SHR taixi,30 min systolic and diastolic pressure had the best antihypertensive effect,better than 60 and 90 min.After treatment,the 24 h systolic blood pressure and diastolic blood pressure had better antihypertensive effect,better than 48 and 72 h,indicating that the acupuncture at taixi point has a significant antihypertensive effect.2.Immediately after acupuncture,PET-CT imaging showed that taixi acupuncture could reduce abnormally elevated glucose metabolism in the hypothalamus,thalamus,dorsal thalamus and other regions of SHR compared with normal WKY rats;Compared with the group,taixi acupuncture could reduce the glucose metabolism in the medulla oblongata,suggesting that the hypothalamus,thalamus,dorsal thalamus and medulla oblongata may play a key role in the transient hypotensive effect of acupuncture taixi.After acupuncture treatment,PET-CT imaging showed that acupuncturing taixi acupoint could reduce abnormally elevated glucose metabolism in the thalamus of SHR compared with normal WKY rats;compared with non-acupoint group,acupuncture at taixi could be reduced.Glucose metabolism in the medulla oblongata suggested that the thalamus and medulla may play an important role in the long-term hypotensive effect of acupuncture in taixi.It can be seen that the effect of acupuncture treatment was consistent with the control of the immediate effect of acupuncture.Acupuncture had different selection of acupoints,different blood pressure reduction effects,and different imaging in the brain area,which objectively reflected the acupoint specificity and acupoint-brain correlation theory.Acupuncture and PET-CT imaging after acupuncture showed that the same areas with decreased glucose metabolism before and after SHR group:motor cortex,sensory cortex,and visual cortex;compared with normal WKY group,compared with SHR group,acupuncture the same areas with decreased glucose metabolism before and after taixi group:thalamus,nucleus and striatum.Compared with non-acupuncture points,the same areas with reduced glucose metabolism before and after acupuncturing taixi group:medulla oblongata and pons bridge.Before and after the existence of the same area of ??reduced glucose metabolism,these areas may have immediate blood pressure effect and after treatment,which may existed in the new blood pressure key brain areas,pending further research. |
