The Clinical And Antioxidative Mechanicim Study On Jin's Three Needling In Treating Children's Mental Retardation

The prevention of Mental Retardation(hereinafter,the MR)is thought to be a worldwide medical problem.In modern medicine,researches on MR are focused on the etiology,symptomatic treatment,education and training,with initiative and radical treatment still lagging behind.Acupuncture and moxibustion of the Chinese medicine are thought to have unique cognition of MR,as well as ingenious treatment and rich experience on it.Besides,In recent years,a large number of clinical practices have turned out satisfactory.ObjectiveThis article combines the progress of modern research,from a perspective of clinical research and experimental studies,to explain a more comprehensive and systematic study of the clinical efficacy and mechanism of acupuncture treatment of children with mental retardation.Methods1.Clinical research(1)Clinical efficacy of acupuncture and moxibustion in the treatment of MR: with specific head acupuncture points: "Sishenzhen","Zhisanzhen","naosanzhen" and "niesanzhen" as the main points,addition and subtraction of points were decided upon the traditional Chinese medicine syndrome differentiation,and acupoint injection was also combined.Four months for a course of treatment,218 cases of MR treatment achieved a satisfactory total effective rate of 80.73%.The ten clinical observations,including poor memory and poor calculation power,all showed significant improvement.Among them,limb weakness,convulsions,and salivation improved significantly,with respective efficiencies of 90%,88%,and 73.71%.At the same time,it was found that the effect of acupuncture plus acupoint injection was better than simple acupuncture group,and the effect was still closely related to age,condition,and course of treatment.The younger,milder disease and longer duration oftreatment had better curative effect,which had no obvious relevance with gender and syndrome differentiation.This therapy for MR is the crystallization of Professor Rui Jin's decades of clinical wisdom,which has the effect of adjusting cerebral qi,brewing mentality,supplementing liver and kidney,filling essence,maintaining heart and spleen and promoting the development of brain in MR patients,as well as improve their Intelligence and obstructed adaptive behavior.The influence of acupuncture on IQ of MR patients: Comparing the semplice acupuncture group with the control group(no acupuncture group),with the DME clinical design requirements strictly followed,to test the IQ changes of the two groups(60 cases each)before and after four months..The results showed that the improvement rate of IQ,VIQ,PIQ,total IQ,and IQ of the acupuncture group were notably higher than those of the control group,and the difference was statistically significant.Point: acupuncture can treat MR and improve the IQ of patients.Effects of acupuncture on trace elements in MR patients' hair: modern studies have shown that Zn(Zn),Cu(Cu),Fe(Iron),Cd(Cadmium)and other trace elements have a non-negligible impact on the development of children's nervous system.The genesis of MR is closely related to the abnormality of these elements.We set two groups,with 52 patients in the MR group and 50 healthy persons in the normal control group.The content of seven trace elements such as Zn,Cu,and Fe was detected by hair samples,and changes in elements before and after the acupuncture treatment in the MR group were performed.The results showed that: compared with the normal group,the MR group had low Zn,Cu,Fe content and high Cd content,but there was no significant difference in the contents of Mn,Pb,and Se;after the acupuncture treatment,the contents of Zn,Cu,and Fe increased significantly and the content of Cd decreased significantly in the MR group,compared with before,and the efficacy of acupuncture was closely related to the above-mentioned elemental changes.Point: there are abnormalities in trace elements in patients with MR,and acupuncture treatment has the effect of normalizing the abnormalities of trace elements in the body.The normal coordination of trace elements plays an important role in promoting RNA and protein synthesis in brain as well as the normal conduction of axonal nerves and in the brain and hippocampus,and enhancing SOD activity.Effects of acupuncture on plasma LPO,GSH-Px and erythrocyte SOD in MR:Oxygen radicals and the secondary lipid per-oxidation is one of the important mechanisms leading to brain damage.52 cases of MR patients and 30 cases of normal controls were tested for these three indicators and observed on the effect of acupuncture.The results showed that compared with the normal group,the LPO activity in MR patients increased,but the activity of GSH-Px and SOD decreased;the degree of severity was positively correlated with the increase of LPO,and negatively correlated with the decrease of GSH-Px and SOD;acupuncture treatment could significantly improve GSH-Px and SOD activity and decrease LPO activity.There was a notable difference between before and after the treatment.Point: there are obvious lipid per-oxidation injury and insufficient activity of antioxidative enzymes in MR patients.Acupuncture treatment of MR is closely related to its ability of enhancing antioxidative capacity of the body,inhibiting free radical oxidation system,and harmonizing the balance between oxidation and antioxidation.2.animal experimentationTo further explore the effect and mechanism of acupuncture,we selected animal models of learning and memory impairment induced by electroshock in rats,to directly observe the effects of acupuncture on lipid per-oxidation,nucleic acid protein synthesis,and learning and memory function in animal brains.(1)The influence of acupuncture on the learning and memory of model animals:The staggered labyrinth test was used to observe the learning and memory status of the electroshock group,acupuncture group and normal group.The results showed that acupuncture can significantly reduce the number of times that the electroshock rat reached the learning standard and improve the accuracy of the memory experiment.When the electroconvulsion was stopped,the acupuncture group recovered faster than the electroconvulsive group,which proved that the acupuncture could improve the electroconvulsion-induced learning and memory impairment,and rehabilitate the damaged learning and memory ability as soon as possible.(2)Effects of acupuncture on the activity of LP0,GSH-Px and SOD in brain tissue: Compared with normal group,the activity of LPO in brain tissue was significantly increased while the activity of SOD and GSH-Px were significantly decreased in the electroshock group;compared with theelectroshock group,the level of LP0 in the brain was significantly decreased while activities of SOD and GSH-Px were significantly increased in the acupuncture group,indicating that acupuncture can enhance the activity of antioxidative enzymes in the brain to a certain extent,eliminate LP0,and reduce the damage of brain tissue caused by lipid per-oxidation.It is believed to be one of the mechanisms by which acupuncture can improve learning and memory abilities of rats and treat MR.(3)Effects of acupuncture on RNA and protein synthesis in brain tissue:The effect of acupuncture on RNA and protein synthesis in hippocampus and cerebral cortex was studied by means of radioactive incorporation of[3H]uridine [3H]leucine.The results showed that in the electroshock group,[3H]uridine and [3H]leucine intake decreased in the hippocampus and cerebral cortex,and synthesis of RNA and protein was impaired,resulting in learning and memory impairment in rats.Acupuncture increased the content of RNA and protein in the hippocampus and cerebral cortex,promoted the synthesis of RNA and protein,and improved the ability of learning and memory in rats.It was speculated to be one of the mechanisms of clinical acupuncture treatment for improving IQ.ResultsThe clinical curative effect analysis showed that combined with TCM syndrome differentiation and treatment,218 cases of the disease were treated with the syndrome plus or minus,the total effective rate was 83.73%,and ten clinical observation indexes of poor memory were obviously improved.At the same time,the curative effect of acupuncture plus point injection was more obvious than that of simple acupuncture.The curative effect of acupuncture and acupuncture and the age and the condition of the disease were more obvious.It is closely related to the course of treatment,but has no obvious correlation with gender and syndrome differentiation.In the IQ test,60 cases of MR patients were treated with pure acupuncture and the control of blank control(that is no acupuncture group).The results showed that acupuncture could obviously improve the IQ of MR patients and showed the progressive rate of IQ.The changes of language IQ,IQ,total IQ and IQ class were significantly higher than those of the control group.The change of trace elements showed that MR patients had abnormal trace elements in the body,showing low Zn,low Cu,low Fe and high Cd.Acupuncturecould obviously improve the trace element conductance in the body of MR patients,which was lower than that of normal control group,which was higher than that of the control group,and the effect of acupuncture and the trace element spectrum were higher than that of the control group.Normalization is positively correlated,indicating that adjusting microelement abnormality is one of the mechanisms of acupuncture treatment of MR.The determination of LPO and GSH-Px in plasma and the detection of SOD in erythrocyte: the activity of LPO in plasma of children with MR was significantly higher than that of normal children;the activity of GSH-Px and erythrocyte SOD in the plasma of children with MR was lower than that of normal children,and the severity of the disease was positively correlated with the lipid peroxidation in the body,and was associated with the activity of antioxidant enzymes.Acupuncture can obviously improve the activity of GSH-Px and SOD and reduce the activity of LPO.It shows that acupuncture treatment of MR is closely related to the anti oxidation ability of the body,the inhibition of free radical oxidation system,and the balance of oxidation-the balance between oxidation and oxidation.The learning and memory impairment model of rats induced by electroshock was selected and the maid labyrinth test was carried out.It was observed that acupuncture could significantly reduce the number of test times of the electroconvulsive rats,improve the correct rate in the memory experiment,and promote the recovery of the impaired learning and memory ability as soon as possible.It is shown that acupuncture can improve the learning record of electroshock.Recalling a defect of a part of intelligence.Compared with the normal group,the activity of LPO in the brain tissue of electroshock rats increased significantly and the activity of SOD and GSH-Px decreased.The brain tissue of electroconvulsive rats had obvious lipid peroxidation damage.At the same time,the acupuncture treatment of Jin three needles could significantly increase the activity of GSH-Px and SOD in the brain tissue of MR animals.It also indicated that acupuncture treatment could increase the clearance of LPO and thus reduce the clearance of LPO.A large number of oxygen free radicals produced by brain injury lead to lipid peroxidation damage.Protein and RNA are closely related to learning and memory.Hippocampus and cerebral cortex are the main parts of learning and memory.The proteinand RNA synthesis of hippocampal and cerebral cortex in electroconvulsive rats decreased significantly,which showed that [3H] uridine and PH] leucine intake were reduced,while acupuncture could significantly increase the absorption rate of [3H] uridine and [3H] leucine in the hippocampus and cerebral cortex of MR animals,and improve the anti injury ability of the above brain regions.In order to antagonism and improve the damage and influence of electroconvulsive shock on RNA and protein synthesis,acupuncture can improve the learning and memory ability of rats through the above ways.ConclusionThis study shows that acupuncture has certain effect to treat MR.It has unique advantages in treating clinical symptoms and improving IQ.As for the mechanism,this text suggests that it functions through the role of meridian acupoints to adjust the yin and yang,make up for its deficiency and diarrhea its excess,and thus plays a positive role in integration,improves the abnormality of trace elements,reduces the lipid per-oxidation damage in the brain and promotes brain RNA and protein Synthesis,which consequently promotes the improvement of clinical symptoms and recovery of mental function.
