Objectives:To study the level of serum PGC-1?/FNDC5/BDNF signal pathway and the thioredoxin system(Trx1/Trx2/Trx R1/Trx R2)which would be relevant to the clinical features in patients with bipolar depressive disorder and major depressive disorder,and to explore the correlation between PGC-1?/FNDC5/BDNF signal pathway and thioredoxin system which may play a part in the pathogenesis of bipolar depressive disorder and major depressive disorder.Methods:26 patients with bipolar depressive disorder(BDD),20 patients with major depressive disorder who met the DSM-5 diagnostic criteria for bipolar depressive episode and major depressive disorder and 28 healthy controls(NC)were recruited in the study through a strict structural clinical interview.Then the subjects were investigated by the General Situation Questionnaire,Hamilton Depression Scale(HAMD),Hamilton Anxiety Scale(HAMA),Symptom Check List 90 and Pittsburgh Sleep Quality Index(PSQI).Polysomnography were used to collect objective sleep parameters of subjects.After recruiting,4ml morning fasting blood of all subjects were taken from left cubital vein between 7:30am and 8:30 am,then the serum samples were collected by centrifugal separation(4?3000r/m).The level of serum PGC-1?,FNDC5,BDNF,Trx1,Trx2,Trx R1,Trx R2 were detected by enzyme-linked immunesorbent assay(ELISA).The SPSS21.0analysis software was used to record a data base and perform the statistic analysis.Results:1.Campared with MAA group,the symptoms of sensitive interpersonal relationship,depression,phobia,paranoid,psychosis in MDD group were more serious(P<0.05).2.The PSQI total score,sleep quality factor score,sleep latency factor score,sleep efficiency factor score,sleep disorder factor and day time factor score were significantly different among BDD group,MDD group and HC group(P<0.05).But there was no difference in the PSQI scores between BDD group and MDD group(P>0.05)3.The polysomnography data of total sleep time,sleep efficiency,wake time after sleep onset,sleep latency and REM latency were significantly different among BDD group,MDD group and HC group(P<0.05).But there was no difference in the PSG parameters between BDD group and MDD group(P>0.05).4.The level of serum PGC-1?,FNDC5,BDNF,Trx R1,Trx R2,Trx1 and Trx2 were significantly different among BDD group,MDD group and HC group(P<0.05).Besides,the level of serum BDNF in BDD group was much lower than that in MDD group(P<0.05).5.There was a significantly correlation between the serum PGC-1?/FNDC5/BDNF signal pathway and the thioredoxin system(Trx1/Trx2/Trx R1/Trx R2)(P<0.05).6.PGC-1? and FNDC5 were risk factors for patients with bipolar depressive disorder and major depressive disorder,BDNF,Trx R1,Trx R2,Trx1 and Trx2 were protective factors for patients with bipolar depressive disorder,and Trx R1,Trx R2 and Trx1 were protective factors for patients with major depressive disorder(P<0.05).7.Each matter of the PGC-1?/FNDC5/BDNF signal pathway and thioredoxin system(Trx1/Trx2/Trx R1/Trx R2)was significant correlatted with scores or parameters of HAMD,HAMA,SCL-90,PSQI and PSG(P<0.05).Conclusions:1.The level of serum PGC-1?,FNDC5,BDNF,Trx R1,Trx R2,Trx1 and Trx2 in patients with bipolar depressive disorder were abnormal,and the level of serum PGC-1?,Trx R1,Trx R2,Trx1 in patients with major depressive disorder were abnormal,which may provide a reference for biomarker of bipolar depressive disorder and major depressive disorder.2.Serum BDNF may be a biomarker which indicate a distinction between bipolar depressive disorder and major depressive disorder.3.The PGC-1?/FNDC5/BDNF signal pathway in patients with bipolar depressive disorder and major depressive disorder not only correlates with thioredoxin system(Trx1/Trx2/Trx R1/Trx R2),but also influences it.4.The PGC-1?/FNDC5/BDNF signal pathway and thioredoxin system which may involve in pathogenesis of bipolar depressive disorder and major depressive disorder are correlated with symptoms of depression,anxiety,subjective and objective sleep.
