| Post Stroke Depression(PSD)is a kind of affective disorders which has clinical features of depression,decreased activity,slow thinking and cognitive function.It mainly displays in extending length of hospitalization,higher risk of dying,complications of treatment for the body disease,bring great burden to family and society.Moreover,the PSD is also independent risk factor for recurrence of stroke.Clinical reports at home and abroad indicates that incidence rate of PSD is 25%to 80%.The current domestic studies on PSD are mostly in the field of drug therapies.And it has the characteristic of the side effects,corresponding to adapt to the indications and contraindications,and high recurrence rate,etc.We has obtained the satisfactory curative effect and overcome the deficiency and limitation of western medicine by used Mongolian medical acupuncture treatment of the disease in recent years,which is simple and no side effects.The mechanism of the method is unknown and most patients still dependent on drug,and application is limited by a lot of reasons.It is very necessary to understand its mechanism.ObjectivesExplore Mongolian medical acupuncture treatment on brain function and mechanism of poststroke depression model rats.Methods1.Grouping:the SPF male SD rats,weight during 280 g to 300 g.One week after adaptive feeding,we selected weight and sugar water consumption of similar 44 before experiment rats,there were eight rats in the sham operation group,and other rats were randomly divided into Mongolian medical acupuncture group,model group,fluoxetine hydrochloride to ensure at least eight rats in each group.2.Preparation and behavior model of observation:Used the model preparation methods such as Longa and modified production focal cerebral ischemia reperfusion animal models.To prevent postoperative infection by intraperitoneal injection of three consecutive days 40000 u of penicillin.Rats were given points after wake up,according to Bederson score with neurologic deficits(grade 1 or higher)into the group of rats.The eighth day of Postoperative chronic unpredictable mild stimulation was given combined with solitary(except the sham operation group).Random every day to give a stimulus,a kind of stimulation can’t continuous use for 14 days.3.Intervention:after the stress of Mongolian medical acupuncture group used acupuncture therapy on Mongolian medical Dinghui-acupoint and frontal acupuncture point,about 0.2 mm or so depth,retaining needle 20 min every time,once daily.Fluoxetine hydrochloride group match into fluoxetine suspension with distilled water,according to 4.06 mg/kg by the stomach,once daily.Model group by gastric perfusion of distilled water.Sham operation group without any intervention.Intervention time 14 days.4.The materials and test:after the experiment,by intraperitoneal injection of 10%chloral hydrate anesthesia.Then open rats blood from the abdominal aorta after centrifugation on serum under test.Rapid brain,blood after the break the ice tray,stripped the hippocampus and cortex in a frozen pipe,using liquid nitrogen quick frozen and-80℃refrigerator under test.With ELISA detection technology,according to the test kit instructions serum cytokines and sodium element,the hippocampus nerve monoamine transmitter levels of the organization.Do after internal fixation of rat body harden beheaded brain in 4%paraformaldehyde under test,with immunohistochemical detecting technology in brain tissue of the expression of BDNF and TrKB.5.Statistical processing with SPSS 19.00 statistical software for statistical analysis.All the data mean standard deviation(X± s),the comparison between groups using single factor analysis of variance,there are significant difference in P<0.05.Results1.The effect of Mongolian medical acupuncture on PSD model rats behavior:1.1 There was no significant difference(P st13 0.05)in rats weight of each group before building.Building after compared with Sham operation group,model group,the Mongolian medical acupuncture group,fluoxetine hydrochloride group three slow weight gain(P<0.05).After treatment compared with Sham operation group,model group rats weight gain obvious difference(P<0.01).Compared with model group,Mongolian medical acupuncture group and the fluoxetine hydrochloride group rats weight increased significantly(P<0.05).Mongolian medical acupuncture group and there was no significant difference between the fluoxetine hydrochloride group(P>0.05).1.2 each group rats sugar water consumption in front of the building there was no significant difference(P>0.05).Building after compared with Sham operation group,model group,the Mongolian medicine acupuncture group,the fluoxetine hydrochloride group three group of sugar water consumption decrease(P<0.01).After treatment compared with Sham operation group,model group rats sugar water consumption is decreased obviously(P<0.01).Compared with model group,Mongolian medicine acupuncture group and the fluoxetine hydrochloride group rats sugar water consumption increased significantly(P<0.05).Mongolian medicine acupuncture group and the fluoxetine hydrochloride group there was no significant difference(P st 13 0.05).1.3 open box experiment after intervention.compared with control group.model group rats activity time and the number of vertical movement were significantly decreased(P<0.01).Compared with model group.Mongolian medicine acupuncture group and the fluoxetine hydrochloride group rats activity time and the number of vertical movement were significantly increased(P<0.05).Mongolian medicine acupuncture group and the fluoxetine hydrochloride group there was no significant difference(P>0.05).2.The influence of Mongolian medical acupuncture on PSD model rats serum levels of cytokines and sodium element:2.1 the influence of serum IL-1 beta,IL-2,IL-6:compared with control group,model group rats serum IL-1 beta,IL-2,IL-6 levels increased significantly(P<0.01,or P<0.05).Compared with model group,Mongolian medicine acupuncture group and the fluoxetine hydrochloride group rats serum IL-2,IL-6 content significantly decreased(P<0.01,or P<0.05).IL-1 beta content was decreased,but no statistical difference(P>0.05).The comparison between Mongolian medical acupuncture group and fluoxetine hydrochloride group was no significant difference(P>0.05).2.2 the influence of the serum levels of IFN-a,IFN-y,TNF-a:compared with sham operation group,model group rats serum.TNF-a,IFN-a content increased significantly(P<0.01);Can not influence the content of IFN-γ(P>0.05).Compared with model group,Mongolian medical acupuncture group and the fluoxetine hydrochloride group rats serum,TNF-a,IFN-α content significantly decreased(P<0.01,or P<0.05),IFN-γcontent was decreased but without statistical significance(P>0.05);Mongolian medical acupuncture of IFN-y content had no effect.Between Mongolian medical acupuncture group and the fluoxetine hydrochloride group was no significant difference(P>0.05).2.3 The effect of serum sodium element(ANP):compared with control group,model group rats serum ANP levels increased significantly(P<0.01).Compared with model group,Mongolian medicine acupuncture group and the fluoxetine hydrochloride group rats serum ANP content was decreased,but no statistical significance(P>0.05).Between Mongolian medical acupuncture group and the fluoxetine hydrochloride group was no significant difference(P>0.05).3.The effect of Mongolian medical acupuncture on PSD model of rat hippocampus neural monoamine neurotransmitter content.Compared with sham operation group,model group rats hippocampus DA,NE,5-HT content reduce(P<0.01);Mongolian medical acupuncture group compared with model group and the fluoxetine hydrochloride group in the rat hippocampus DA,NE,5-HT content increased significantly(P<0.01,or P<0.05).The comparison between Mongolian medical acupuncture group and the fluoxetine hydrochloride group,there was no significant difference(P>0.05).4.The Mongolian medical acupuncture on PSD model rats brain tissue of the brain derived neurotrophic factor BDNF and TrKB expression effect.Model group compared with sham operation group in the rat hippocampus BDNF and TrKB expression(P<0.05);Mongolian medical acupuncture group compared with model group and the fluoxetine hydrochloride group of BDNF and TrKB expression increased but without statistical significance(P>0.05);The comparison between Mongolian medical acupuncture group and the fluoxetine hydrochloride group,there was no significant difference(P stl3 0.05).Model group compared with control group rats cortex BDNF and TrKB reduce(P<0.05);Mongolian medicine acupuncture group compared with model group and the fluoxetine hydrochloride group of BDNF and TrKB expression has no statistical significance(P>0.05).Conclusion1.Manufacturing after cerebral ischemia reperfusion model used tail,oscillation.black and white reversed,fasting ban,bound water,damp bedding stimulus such as increase the psychological pressure to produce the PSD model.Build model rats after depression,dark color,slow weight gain,activity decreased significantly,the phenomenon such as eating less.Made mold method and model of rats after various indicators in accord with physical and mental features of medical excessive and three root seven disorders,affect "Badagan Hei"caused PSD.2.PSD model rats serum cytokines(ANP)and sodium element observation indexes changed in different degree.These indicators changed directly influenced the occurrence of PSD.From the change of ANP can get affected by heart.Brain damage affects the heart,and the heart and brain are connected by two Hector in accordance with the foliage of Mongolian medicine basic theory.Therefore,the "heart" and "brain" is the lesion site of PSD.3.The experimental results show that Mongolian medical acupuncture regulating PSD rat serum cytokines,hippocampal neural monoamine neurotransmitter and brain tissue of brain derived neurotrophic factor and receptor TrKB expression.Which fully embody the Mongolian medicine acupuncture regulation of three root seven disorders,unblocked blood running,repair damaged brain and white veins,anti-inflammatory,and change the internal energy of relevant system,regulate the body’s biological information to treat the PSD. |
