ObjectiveThis topic in Mongolian medical for PCPA cause insomnia rats model as the research object,it observe the behavior changes of rats,brain tissue pathological morphological changes,cell factor beta IL-1?,IL-2,IL-6,TNF-a and neurotransmitters 5-HT,GABA content,the influence of Mongolian medical warm acupuncture combined with Su Gemu le-3 for the treatment of insomnia rats.Methods1.70 SD rats were randomly divided into 7 groups,the normal group,model group,Su Gemu le-3 high dose group,Su Gemu le-3 middle dose group,Su Gemu le-3 low dose group,warm acupuncture combined with Su Gemu le-3,10rats in each group.The mouse model was used to replicate the insomnia rat model with intraperitoneal injection of chlorphenylalanine.2.HE staining was performed in the cerebral cortex,hypothalamus and hippocampal slices,and the pathological changes were observed in the light microscope.3.The contents of IL-1?,IL-2,IL-6 and TNF-a in rats were detected by ELISA.4.High performance liquid chromatography(HPLC)was used to detect the changes in the contents of neurotransmitters 5-HT in rats and the content of GABA in the hypothalamus.Results1.Weight change:before PCPA modeling,there was no significant difference in weight between the groups(P>0.05).Before treatment,the weight of rats in the model group was lower than that in the blank group,with significant difference.The weight of the rats in the model group was lower than that in the blank group,and the weight of the rats in the high-dose group was higher than that in the model group,and there was a significant difference.2.Behavior:in the tail test,there was no significant difference in the movement time between the groups before PCPA formation(P>0.05).Before treatment,the rats in the model group had greater movement time than the blank group,with significant difference.The rats in the former model group were more active than the blank group,and the time of the rats in the high-dose group was lower than that in the model group,and there were significant differences.In forced swimming,there was no significant difference between the head of the rats in the head of each group(P>0.05)before the PCPA model was formed.Before treatment,the head of the model group was submerged in water for less than the blank group,with significant difference.The head of rats in the former model group was submerged in water less than the blank group,and the head of the rats in the high-dose group of sugule-3 was flooded with water over the model group,and there was a significant difference.In the open field experiment,the activity time of the pre-material model group was greater than that of the blank group(P<0.01).There was no significant difference(P>0.05)in the model group.In the vertical activity,the rats in the model group stood more than the blank group,with significant difference.3.Brain histopathological changes:the combination of Mongolian medicine temperature needle and sugumo-3 soup improved the morphological changes of rat brain.4.The content of IL-1?:the combination of Mongolian medical warm acupuncture and sugemule-3 decoction could reduce the level of serum IL-1? in rats with insomnia model,which was significantly different from the model group(P<0.01).The content of IL-2:the combination of Mongolian medicine warm acupuncture and sugemule-3 decoction could reduce the serum IL-2 content in the rats with insomnia model,which was significantly different from the model group(P<0.01).The content of IL-6:the serum IL-6 content of the rats with insomnia model can be reduced by the combination of the Mongolian medical warm acupuncture and the sugemule-3 soup,which is significantly different from the model group(P<0.01).TNF-a content:the serum TNF-a content of the rats with insomnia model was reduced by the combination of the Mongolian medical warm acupuncture and the sugemule-3 decoction,which was significantly different from the model group(P<0.01).5.5-HT content:the serum 5-HT content of the rats with insomnia model was improved by the combination of the Mongolian medical warm acupuncture and the sugarmule-3 soup,which was significantly different from the model group(P<0.01).GABA content:the combination of Mongolian medical warm acupuncture and sugumule-3 soup can improve the content of GABA in the hypothalamus of rats with insomnia model,which is significantly different from the model group(P<0.01).Conclusion1.The combination of Mongolian medical warm acupuncture and sugemule-3 soup can improve the mental state and behavioral ability of rats with insomnia model.2.The combination of Mongolian medical warm acupuncture and sugemule-3 soup has the effect of improving the pathological changes of cerebral cortex,hypothalamus and hippocampal neurons in rats with insomnia model.3.The combination of Mongolian medical warm acupuncture and sugemule-3 soup improves sleep by improving the levels of serum IL-13,IL-2,IL-6 and TNF-a in rats with insomnia model.4.The combination of Mongolian medical warm acupuncture and sugemule-3 soup improved sleep by improving the serum 5-HT and GABA content in the rats with insomnia model.
