1.BackgroundOsteoarthritis(OA)is a common degenerative joint disease,involving the entire synovial joint,including cartilage,synovial membrane and subchondral bone.Its main clinical manifestation is joint pain.It not only leads to the limitation of joint function and the decline of quality of life,but also leads to the mobility inconvenience of the elderly.Osteoarthritis pain affects about 70 percent of people aged 55 to 70 in western countries.Currently,the most effective treatment for osteoarthritis is joint surface replacement.Chondroitin sulfate(CS)is an important component of the extracellular matrix of cartilage and has been widely used as one of the main drugs and health care products related to joint pain.Chondroitin sulfate has obvious cartilage properties and preferentially enters cartilage tissue to protect cartilage.The main sources of chondroitin sulfate are cartilage of animals such as poultry,livestock and sharks,but poultry and livestock have low cartilage content,and sharks are national wild protected animals with relatively scarce resources.At present,the development of China's squid breeding industry is in good condition.Sturgeon bone(SB)is one of the main wastes produced during the further deep processing of carp in fishery production,and its cartilage.The chondroitin content is quite high.Studies have shown that the chemical structure of chondroitin sulfate in squid is different from chondroitin sulfate in other animal bones,and there are few reports on the activity and mechanism of action of chondroitin sulfate.In this study,an animal model of osteoarthritis in rats was induced by using monosodium iodoacetate(MIA)to investigate the effect of chondroitin sulfate in the treatment of osteoarthritis induced by sodium iodide in rats.The activity and mechanism of action of chondroitin sulfate for the treatment of osteoarthritis.2.ObjectivesThe purpose of this study is to evaluate and further study the therapeutic effect and mechanism of squid bone chondroitin sulfate(CSSB)on sodium iodide(MIA)-induced osteoarthritis(OA).The potential medicinal value of squid bone chondroitin sulfate in the treatment of osteoarthritis.3.MethodsThe present study was evaluated the therapeutic effect of CS from SB(CSSB)onmonosodiumiodoacetate(MIA)-induced osteoarthritis(OA)pain and further explored the potential medicinal value of CSSB.The OA pain model was induced by intra-articular injection of MIA and then treated with CSSB.The results showed that.on the organismic level,CSSB can significantly reduce the joint swelling,reduce the pathological injury of the joints,decrease the levels of IL-1,TNF-a and PGE2 in synovial fluid,revers of hind paw support and paw withdrawal mechanical threshold decreased caused by MIA.In addition,mechanistically at the molecular level,these effects are elicited via down-regulation of the protein levels of down-regulate the protein expression of matrix metalloproteinase MMP-1,MMP-3 and up-regulate the protein expression of TIMP-1.4.ResultsThe above experimental results indicate that squid bone chondroitin sulfate(CSSB)can alleviate cartilage degradation by regulating matrix metalloproteinases(MMPs)and matrix metalloproteinase tissue inhibitors(TIMPs)to alleviate sodium iodide(MIA)Animals with osteoarthritis pain.5.ConclusionsTherefore,squid bone chondroitin sulfate can be used as a potential pharmaceutical ingredient.It has potential therapeutic effects on osteoarthritis induced by sodium iodoacetate,and can further expand the social and economic value of squid bone chondroitin sulfate.
