Objective To observe effect of supervisor's prescription,"He Chang Yin",on the signal pathway of NLRP3,NLRP6 inflammation of model rat,(the expression of ASC and caspase 1 and IL-1?,the influence of the content of IL-18,discussed and intestinal drink on ulcerative colitis model rats NLRP3,the influence of the inflammatory corpuscle NLRP6 signaling pathways)and discussing the possible mechanism of "He Chang Yin"on treating UC model rats.Methods 60 healthy male SD rats were randomly divided into 6 groups respectively,the normal group,model group,mesalazine(5-SA)in treatment group,and treatment groups treat with high,medium and low dose of "He Chang Yin",each group is 10.In addition to the normal group,the rest of the 5 groups are establishing the UC rat model induced by(2,4,6-trinitrobenzene sulfonicacid,TNBS,)After drug intervention,to observe the disease activity index(DAI),colon histopathological grading(HPS)By observation of pathological changes in rat colon lesions,immunohistochemical method to detect colon tissue NLRP3,ASC and the expression of caspase 1,real-time fluorescent quantitative PCR(qRT-PCR)method to detect NLRP3,NLRP6,ASC and caspase 1 mRNA expression,ELISA method to detect the serum IL-1?,IL-18 content.Results 1.DAI rats score showed that compared with normal rats,the rats in the model group the DAI score increased significantly,the difference was statistically significant(P<0.01);compared with the model group rats,and intestinal Decoction high dose group and Western medicine group rats DAI score decreased significantly,the differences were statistically significant(P<0.01),and He Chang Yin in low and medium dose group rats DAI scores were lower than the model group,the difference was statistically significant(P<0.05);there was no significant difference in DAI score between the treatment groups(P>0.05)2.Each rat colon tissue pathology score showed that compared with normal rats,the pathological colon tissue of the rats in the model group was significantly increased,the difference was statistically significant(P<0.01);compared with the model group rats score significantly decreased and intestinal He Chang Yin and high dose group and Western medicine group colon tissue pathology in rats,and the difference was statistically significant(P<0.05),intestinal and colonic pathological Decoction low dose group of rats was also lower than the model group,but the difference was not statistically significant(P>0.05);the difference between the treatment group colon histopathology score was not statistically significant(P>0.05)3.The expression of IL-1?:compared with normal rats,the content of serum IL-1? in the model group was significantly increased,the difference was statistically significant(P<0.01),compared with the model group rats,and He Chang Yin low and medium dose group and Western medicine group rats serum IL-1? content decreased,and the difference has statistical significance(P<0.05),and the serum IL-1? content of He Chang Yin in high dose group decreased significantly,and the difference was statistically significant(P<0.01),there was no significant difference between the content of serum IL-1?treatment group(P>0.05).4.The expression of IL-18:compared with normal rats,the serum IL-18 of the rats in model group were significantly increased,the difference was statistically significant(P<0.01),compared with the model group rats,and He Chang Yin low,medium and high dose group and Western medicine group rats serum IL-18 content decreased significantly,and the difference was significant(P<0.01),there was no significant difference between the content of IL-18 in serum of each treatment group(P>0.05).5.The expression of Caspase-1 mRNA:compared with normal rats,the increased expression of Caspase-1 in rat model of colon tissue group mRNA,the difference was statistically significant(P<0.05),compared with the model group rats,and He Chang Yin medium and high dose group and Western medicine group colon tissue of rats Caspase-1 expression of mRNA decreased,and the difference was significant the difference(P<0.05),and He Chang Yin low dose group rats Caspase-1 mRNA expression compared with the model group also decreased,but the difference was not statistically significant(P>0.05),the treatment group Caspase-1 mRNA expression showed no significant difference(P>0.05)6.The expression of ASC mRNA:compared with normal rats,the increased expression of ASC in rat model of colon tissue group mRNA,the difference was statistically significant(P<0.05),compared with the model group rats,and He Chang Yin low in the high dose group and Western medicine group colon tissue of rat ASC mRNA expression decreased and the difference was statistically significant(P<0.05),the treatment group ASC mRNA expression showed no significant difference(P>0.05).7 expression of NLRP3 mRNA:compared with normal rats,the expression model of colon tissue of rats in NLRP3 group mRNA increased significantly,the difference was statistically significant(P<0.01),compared with model group,high dose of colon tissue and He Chang Yin group and Western medicine group rats NLRP3 mRNA expression was significantly decreased,with statistically significant differences(P<0.01),medium dosage group and intestinal NLRP3 expression of mRNA decreased,the difference was statistically significant(P<0.05),and He Chang Yin low dose group rats NLRP3 mRNA expression compared with the model group also decreased,but the difference was not statistically significant(P>0.05),the treatment group NLRP3 mRNA the expression of the difference was not statistically significant(P>0.05).8 NLRP6 mRNA compared with normal rats,the expression model of colon tissue of rats in NLRP6 group mRNA increased significantly,the difference was statistically significant(P<0.01)compared with the rats in the model group,high dose of colon tissue He Chang Yin n group and Western medicine group rats NLRP6 mRNA expression was significantly decreased,the difference was statistically significant(P<0.01),He Chang Yin medium dosage group NLRP6 expression of mRNA decreased,the difference was statistically significant(P<0.05),He Chang Yin low dose group rats NLRP6 mRNA expression compared with the model group also decreased,but the difference was not statistically significant(P>0.05),the treatment group had no significant difference between the expression of mRNA between NLRP6(P>0.05).Conclusion1.He Chang Yin can improve the disease activity index(DAI)and the histopathological score(HPS)of rats with ulcerative colitis(UC).2.He Chang Yin can inhibit the expression of NLRP3?NLRP6?ASC?caspase-1 and NLRP3?NLRP6?ASC?caspase-1 mRNA,and also can reduce the content of IL-1??IL-18 in serum.3.He Chang Yin may inhibit the expression of IL-1? and IL-18 by inhibiting the NLRP3 and NLRP6 inflammatory signaling pathway,and reduce the damage of colonic mucosa in rats with ulcerative colitis.
