Objective: Chronic nonspecific ulcerative colitis (CNUC) is called ulcerative colitis (UC) in short. It is a kind of Chronic inflammatory disease. The cause of it doesn't definite. The main presentation of it are as follows: intermitent diarrhoea with mucus and blood at stool, epigastric pain, tenesmus and constipation of few patients. Some companied with joint, Skin, eye, mouth, liver and gall disease and so on, It is always lingering, refractory and easy to relapse. In recent years, the research about its etiopathogenesis has made great progress, especially on the mechanism of molecular immunology. So far, we have been lacking of effective remedies for curing/it. In order to find an effective ways to cure it, during the past year, the scholars of home and abroad have been intent on the study of Tradition of Chinese Medicine (TCM). LichangSiFang (LCSF), which includes WuMeiWan (WMW), BaiTouWengTang (BTWT), SenglingBaiShuSan (SLBSS), TongXieYaoFang (TXYF) are Ways of treating UC, Which had some curative effect on UC, But they hadn't been compared in themselves and made conclusive evidence.This subject is about immunoregulatory and treatment effect of LCSF on molecular level by making rat model of UC, especially they were observed the action of UC on cytokine and apoptosis to make a progress for treatment UC with TCM.Methods: In the experiment, ninties eight SD rats were randomly divided into seven groups, Every group had fourteen rats (seven rats were male, seven rats were female), namely the normal control group, the pathologic control group, the solfasalazine (SASP) group, the WMW group, BTWT group, SLBSS group, TXYF group. Except the normal control group, the other six animal model groups of UC were made through the integration of general immunity and local stimulation (DNCB and acetic acid), the six animal model groups of UC were treated by distilled water, solfasalzine, TXYF, WMW, BTWT, SLBSS, TXYF respectively. After these treatment, the changes of symptoms and some immune indexes in the colon tissue such as IL-6, IE-8, TNF-ot, IL-10, TNF-ctmRNA gene expression in the colon tissue, apoptosis in the colon tissue, the pressing protein of Bcl-2, Bax, Fas were observed and detected. The colon pathological changes of every group were also observed under light microscope and transmission electron microscope.Results: (l)The level of IL-6, IL-8 , TNF- a in pathologic group was significantly higher than that in normal group (P<0.01), and the livel of IL-10 was significantly lower than normal (PO.01). The level of IL-6, IL-8, TNF- a in LiChangLiFan, SASP group was obviously lower than that in pathologic group (P<0.01), and the level of IL-10 was obviously higher than pathologic group (P<0.001). The level of IL-6, IL-8, TNF-a in WMW group, BTWT group, SLBSS group was lower than that in SASP group (P<0.05 or P<0.01), and the level of IL-10 was higher than SASP group (P<0.05 or P<0.01). The level of IL-6, IL-8, TNF- a in TXYF was higher than that in SASP group (PO.05), and the level of IL-10 was lower than that in SASP group (P<0.05). The level of IL-6, IL-8, TNF- a in WMWgroup, BTWT group, SLBSS group is lower than that in TXYF group (P<0.05 or P<0.01), the level of IL-10 is higher than that in TXYF group (P<0.05 or P<0.01). The level IL-6, IL-8, TNF- a in WMW group and BTWT group was lower than that in SLBSS (P<0.05 or P<0.01), and the level of IL-10 was higher than that in SLBSS group (P<0.05 or P<0.01). The level of IL-6,IL-8, TNF- a in WMW group was lower than that in BTWT group (P<0.05 or P<0.01),and the level of IL-10 was higher than that in BTWT group (P<0.01). The case number of the TNF-amRNA expression was significantly in creased in pathologic group than in normal group (P<0.01).(2) The degree of the TNF- a mRNA expression (in situ hybridization way) was decreased in WMW than in pathologic group (P<0.05 or P<0.01).
