Objective:Preliminary study on the mechanism of promoting angiogenesis of human adipose derived stem cells after astragalus polysaccharide interventionMethods:The adipose derived stem cells(ADSC)were obtained from liposuction by collagenase digestion and culture.The osteogenic and adipogenic differentiation ability of ADSC of P3 generation was confirmed by Alizarin s and oil red O staining,and the surface differentiation antigen of ADSC was detected by flow cytometry.Different concentrations of Astragalus polysaccharides(APS)were used to interfere with ADSC,and CCK-8 was used to select the best intervention concentration.The coculture system of ADSC and human umbilical vein endothelial cells(HUVEC)was used as the experimental group,and ADSC alone and APS alone as the control group.The formation rate of endothelial tubular structure was evaluated with APS+ADSC intervention as experimental group,ADSC intervention and no intervention as control group.Western blot was used to detect the expression of Ang-1,integrin,VEGF and ERK,p38,JNK in endothelial cells after 40 ug/ml APS intervention.Results:The ADSC could be stably amplified by enzyme digestion,and the staining of Alizarin Red S and oil red O were all positive.The results of flow cytometry showed that CD90+,CD34-.The results of CCK-8 indicated that the best APS intervention concentration was 40ug/ml.In the cell coculture system,the migration rate of endothelial cells and the formation rate of tubular structure after APS intervention in ADSC were significantly higher than those in the control group(P<0.05).After APS intervention for 48 hours,the expression of Ang-1,integrin and VEGF was up-regulated(P<0.05).After APS intervention on ADSC for 24 hours,the expression of p-ERK1/2 and ERK1/2 in endothelial cells was up-regulated,the difference was statistically significant(P<0.05);the expression of p38 and JNK was not statistically significant.The patients with autogenous fat augmentation mammoplasty were divided into Astragalus interv ention group and control group.Astragalus intervention group used astragalus injection for intrav enous drip for one week after operation,and measured various breast indexes three months after operation,suggesting that Astragalus was significantly better than the control group in improving the survival rate of fat transplantation,with statistically significant difference.Conclusion:Astragalus polysaccharide can promote the proliferation of ADSC and activate its angiogenesis function.The expression of this function may be related to the activation of Angl-Tie2-Erk and/or VEGF-VEGFR-ERK signaling pathway.Astragalus was significantly better than the control group in improving the survival rate of fat transplantation,with statisticall y significant difference.
