Ovarian cancer is the second most common and the most lethal gynecologic malignancy in the western world.So far,there is lack of methods recommended for screening and early diagnostics of this disease.As a consequence,and also due to the absence of early warning symptoms,about 70%of cases is diagnosed at an advanced stage and have bad prognosis.This is mostly due to the progress in surgical technology and contemporary regimes of systemic treatment,as well as some new drugs entering the clinic.In chemotherapy,platinum-based agents such as cisplatin,carboplatin and oxaliplatin form complexes through covalent bonding with DNA analysis,causing DNA damage and subsequently promoting cell death and apoptosis.Although the clinical treatment of cisplatin is effective,the occurrence of drug resistance is still an important factor for its limited efficacy in Ovarian cancer.Therefore,it is of great importance to explore the molecular markers involving in the Ovarian cancer chemo-resistance mechanism.Platelet-derived growth factor receptor alpha(PDGFRA)is the receptor of platelet-derived growth factor(PDGF)protein family,located on the surface of cell membrane,belonging to a type of tyrosine kinase receptor.It plays an important role in regulating cell proliferation,differentiation,growth and development.If not regulated properly,it can cause a variety of diseases(such as ovarian cancer,polycystic ovary syndrome)Accumulating evidences demonstrate that the expression of PDGFRA is related to carcinogenesis.PDGFRA was found to be highly expressed in malignant tumors.Taking into account that PDFGRA performs a significant role in tumor metastasis and apoptosis resistance during tumor metastasis,we postulated that the expression of PDGFRA could affect PIK3R1 promote the expression of inhibitors of apoptosis proteins,which may increase the cisplatin drug resistance of Ovarian cancer cells and restrict the cisplatin chemotherapy efficacy of Ovarian cancer.In summary,this research investigated whether PDGFR could regulate the sensitivity of Ovarian cancer cells to cisplatin and explored the underlying molecular mechanisms.
