ObjectiveThe results of clinical trial about docetaxel showed that half of mCRPC patients were not sensitive to docetaxel chemotherapy,and there was no recognized method to detect the sensitivity of docetaxel in mCRPC patients.In this research,SERS peaks related to the docetaxel sensitivity of mCRPC were explored by using serum surface-enhanced Raman spectroscopy,and a predictive model for the docetaxel-sensitivity of mCRPC was established.MethodsThe subjects were mCPRC patients who underwent docetaxel chemotherapy at our center from July 2016 to March 2018.A total of 32patients were included in this study.According to the effect of docetaxel,they were divided into the effective group?n=17?and the ineffective group?n=15?.The correlation between the relevant clinicopathological characteristics,blood biochemical indexes before chemotherapy,ABCB1gene polymorphism and docetaxel sensitivity of mCRPC was analyzed retrospectively.At the same time,64 matched serum Raman spectra were obtained by using surface enhanced Raman spectroscopy of serum before and after the first chemotherapy.By comparing the changes and differences of serum SERS before and after chemotherapy between the effective group and the ineffective group,Raman peaks related to the sensitivity of mCRPC docetaxel were explored.Combined with PCA-LDA analysis,a prediction model for the sensitivity of mCRPC docetaxel was established,and its predictive efficacy was determined by ROC curve and leave-one-out cross validation.ResultsThere were no significant differences in the relevant clinicopathological characteristics,blood biochemical indexes before chemotherapy and ABCB1 gene polymorphism between the effective group and the ineffective group?p>0.05?.The SERS of serum of the effective group before and after chemotherapy showed significant changes at the peak intensity of 683 cm-1,725 cm-1,1,136 cm-11 and 1,202cm-1?p<0.05?.When comparing the differences of serum SERS peaks before chemotherapy between the two groups,significant differences could be found at the peaks of 638 cm-1,810 cm-1,890 cm-1?p<0.05?and1,136 cm-1?p=0.003?.The prediction models of peak 1,363 cm-11 and PCA-LDA based on Raman data were established respectively,and the AUC values of the ROC curves of the two models were 0.80 and 0.75,respectively.The sensitivity and specificity of the prediction models were71%,80%and 65%,73%through the way of leave-one-out cross validation.ConclusionsThere was no significant correlation between clinicopathological characteristics,blood biochemical indexes before chemotherapy,ABCB1gene polymorphism and docetaxel sensitivity of mCRPC.The serum SERS peak 1,136 cm-11 and PCA-LDA model could play a role in predicting the sensitivity of mCRPC.
