Objective :(1)To observe the effect of electroacupuncture at Shenshu point on pain behavior of rats with sciatica and its effect on the expression of inflammatory factors TNF-α,IL-1β,IL-18 and p65.(2)To observe the changes of WNT signaling pathway in the spinal cord of rat model of sciatica;to observe whether the content of neuronal nitric oxide synthase in rats with sciatica has changed,the symptoms of sciatica and WNT signaling pathway in model rats Whether activation is related to nNOS content in the spinal cord;Methods:Experiment 1 Method: 18 healthy male Sprague-Dawley rats were randomly divided into sham operation group,model group and treatment group,with 6 rats in each group.A rat model of chronic crush injury of the sciatic nerve was established.The treatment group began to receive electroacupuncture at Shenshu point on the first day after surgery.Behavioral tests were performed on rats using Von-Fair,and the rats in the three groups were monitored 1 day before surgery,1st day,3rd day,5th day,7th day,9th day,11 th day,13 th day after treatment.The mechanical mechanical contraction changes the pain threshold and statistical analysis of the test results.On the 14 th day,the content of p65 protein in the spinal cord of each group was detected by Western Blot.The serum levels of TNF-α,IL-1β and IL-18 were detected by Elisa.Experiment 2 Methods: Thirty healthy male Sprague-Dawley rats were randomly divided into sham operation group,model group,treatment group,inhibitor group,inhibitor + acupuncture group to establish a rat model of chronic crush injury of sciatic nerve.The treatment group,the inhibitor group,the inhibitor + acupuncture group were treated with electroacupuncture at Shenshu point on the 3rd day after surgery for 13 days.Behavioral testing was performed on rats using Von-Fair,and the rats in the three groups were monitored 1 day before surgery,1st day,3rd day,5th day,7th day,9th day,11 th day,13 th day of treatment.The changes of the foot pain threshold were observed on the 14 th day after operation.The contents of nNOS,β-catenin,c-Myc protein and m RNA in the spinal cord of each group were detected by Western Blot and PCR.Results:(1)Compared with the sham operation group,the model group had a significantly lower mechanical pain threshold(P<0.05),which was statistically significant.Compared with the model group,the mechanical contraction pain threshold was increased in the treatment group(P<0.05).),has statistical significane.The levels of p65,TNF-α,IL-1β and IL-18 in the model group were significantly higher than those in the sham-operated group(P<0.05),which was statistically significant.After treatment,the rats in the treatment group had p65 in the spinal cord.The levels of TNF-α,IL-1β and IL-18 were all decreased(P<0.05),which was statistically significant.(2)Compared with the sham operation group,the levels of β-catenin and wnt3 a protein in the model group were significantly increased(P<0.05),which was statistically significant.After treatment,the nNOS protein in the spinal cord of the treated group was The contents of β-catenin,wnt3 a and c-Myc decreased(P<0.05),which was statistically significant.The β-catenin,wnt3 a protein and m RNA levels in the inhibitor group were significantly lower(P<0.05),which was statistically significant.Compared with the model group,after treatment,the treatment group,the inhibitor group,and the inhibition group The contents of β-catenin,wnt3 a protein and m RNA in the rat model of acupuncture and moxibustion group were significantly decreased(P<0.05),which was statistically significant.Conclusion:(1)Electroacupuncture at Shenshu Point in the treatment of rat model of chronic crush injury of sciatic nerve can increase the threshold of mechanical contraction pain.The expression of p65,TNF-α,IL-1β and IL-18 is increased in the rat model of chronic crush injury of sciatic nerve.(2)β-catenin and wnt3 a protein were significantly increased,and the WNT signaling pathway was activated in the model.Rats in the treatment group can reduce the activity of WNT signaling pathway-related proteins after acupuncture treatment.(3)The content of nNOS protein and m RNA in the spinal cord of rats with chronic sciatic nerve injury was significantly increased.After reducing the content of nNOS in the inhibitor group,the mechanical contraction pain threshold of rats was also significantly improved.After electroacupuncture treatment,the content of nNOS was significantly decreased.In the inhibitor+acupuncture group,the mechanical contraction pain threshold was more obvious than that in the inhibitor group and the treatment group.(4)After acupuncture treatment,injection of nNOS inhibitor,combined with acupuncture treatment + inhibitor,the activity of WNT signaling pathway was significantly reduced,indicating that electroacupuncture treatment can down-regulate the activity of WNT signaling pathway by decreasing the content of nNOS.
