Objective:To investigate whether resveratrol could prevent muscle atrophy of HiLo rats by reducing oxidative stress of skeletal muscle,and also to investigate whether L-arginine could prevent muscle atrophy of HiLo rats by increasing NO of skeletal muscle.Methods:One hundred and twenty adaptively trained SD rats were randomly divided into normoxic sedentary group(C group),norrmlxic training group(E group),hypoxic sedentary group(HC group),HiLo group(HE group),normoxic sedentary resveratrol group(RC group),normoxic training resveratrol group(RE group),hypoxic sedentary resveratrol group(RH group),hypoxic training resveratrol group(RHE group),normoxic sedentary arginine group(AC group),normoxie training arginine group(AE group),hypoxie sedentary arginine group(AH group)and hypoxictraining arginine group(AHE group).A HiLo model was developed to simulate living at 3500m altitude(12.5%oxygen tensin)10h per day while trained under normoxia with moderate intensity(treadmill grade 10%,speed 25m/min,one hour per day,5 days per week).The rats of control groups were provided free access to basic diet and the rats of resveratrol and L-arginine group were given free access of basic diet and resveratrol or L-arginine with gastric infusion for 2weeks.The body composition,muscle weight of extensor digitorum longus and soleus,BUN,total protein(TP),MDA,SOD and NO of skeletal muscle were measured after 2 weeks.mRNA expressing of NF-?B?MURF1?AKT and mTOR of control,resveratrol and arginine groups were evaluated by Real-time qualitative RT-PCR.Results:1.The body weight,muscle weight and TP were significantly decreased in HiLo rats.Resveratrol and arginine could significantly increase the muscle weight and TP of HiLo rats.2.MDA was significantly decreased in HiLo and hypoxic rats,and SOD had no significant diference;Resveratrol could significantly decrease MDA and increase SOD;Arginine could could significantly decrease MDA and BUN,and increase SOD and NO.3.mRNA expressing of Akt was significantly decreased,and mRNA expressing of NF-?B?MuRfl was significantly increased.Resveratrol could significantly increase mRNA expressing of Akt,and significantly decrease mRNA expressing of NF-?B.4.mRNA expressing of Akt/mTOR was significantly decreased.Arginine could significantly increase mRNA expressing of Akt/mTOR.Conclusions:HiLo could decrease weight,muscle and protein synthesis.The intervention of resveratrol could prevent muscle loss by increasing SOD and decreasing oxidative stress of skeletal muscle.Arginine coule decrease catabolism of protein and muscle loss by increasing NO and SOD,decreasing BUN.Resveratrol could decrease mRNA express of NF-?B and increase mRNA express of Akt.Arginine could increase mRNA express of Akt/mTOR.
