Objective: To investigate endogenous repair mechanisms that improve vascular function,we studied the effects of exercise on vascular endothelial function and the transcriptome of endothelial progenitor cells in postmenopausal women through 12 weeks of aerobic intervention.Methods: In experiment one,30 postmenopausal women were screened out by questionnaires,blood parameters,and arterial ultrasound.They were randomly divided into exercise group and control group.Before and after exercise twelve weeks of aerobic exercise,all subjects were measured FMD,NO,ET-1,E2,four lipids,BMI,blood pressure.In experiment two: Before and after exercise,endothelial progenitor cell levels and endothelial progenitor cell function.Before and after exercise,peripheral blood mononuclear cells,were extracted twice and endothelial progenitor cells were cultured in vitro.In experiment three: RNAseq was used to perform transcriptome test and extract analysis data.Bioinformatics methods were used to perform functional annotation and pathway analysis of differential genes.Results:(1)There were no differences between the groups before exercise.In the exercise group,FMD and NO before and after exercise increased(p<0.05),and there was no significant difference in blood lipids,BMI,ET-1,and E2.(2)After 12 weeks of aerobic exercise,The levels of endothelial progenitor cells increased significantly(p<0.05),Peripheral blood mononuclear cells were identified as endothelial progenitor cells after cultrued;Both migration and proliferation of endothelial progenitor cells were significantly increased(p<0.05).(3)253 differential genes were screened,genes that have risen twice are selected,in which 88 genes were upregulated,and 165 genes were down-regulated.(4)Analysis of GO and KEGG data systems revealed that the genes involved in proliferation and migration were mainly located in the cytokine-cytokine receptor-receiving pathway,and 17 genes such as GAS6,MMP7,E2F6,NR2E1,VEGF,IL6,CDK10,Bach1,Bach2,and SOSC3 were related to proliferation and migration.(5)IL-6 and AGT were found in the network of differential gene networks by screening protein interaction networks.Conclusions:(1)12-week exercise intervention can improve endothelial function in postmenopausal women by improving NO levels.(2)12-week aerobic exercise significantly increased the level of endothelial progenitor cells,migration,and proliferation of postmenopausal women.(3)12-week aerobic exercise can increase the proliferation of endothelial progenitor cells and improve endothelial function by upregulating GAS6,MMP7,E2F6,NR2E1,VEGF,and downregulating IL6,CDK10,Bach1,Bach2,and SOSC3.(4)12-week aerobic exercise can upregulate GAS6,CSPG4,VEGF,CXCL12 and downregulate IL6,AGT and ACE,improving endothelial progenitor cell migration function of postmenopausal women,and then improve blood vessel function.
