Synthesis of phthalocyanine photosensitizers for photodynamic therapy studies

Abstract/Summary:
A group of 20 new silicon and aluminum phthalocyanines designed for use as photosensitizers in photodynamic therapy studies has been made and characterized. In addition, one previously known silicon phthalocyanine suitable for use in photodynamic therapy studies has been remade and further characterized. Of these 21 photosensitizers, 18 are related to Pc 4, HOSiPcOSi({dollar}rm CHsb3)sb2(CHsb2)sb3N(CHsb3)sb2{dollar}, a known photosensitizer that exhibits properties desirable in a photodynamic therapy photosensitizer. The remaining three photosensitizers are related to Pc 5, HOSiPcOSi({dollar}rm CHsb3)sb2(CHsb2)sb3Nsp+(CHsb3)sb3{dollar} I{dollar}sp-{dollar}, a known photosensitizer belonging to a class that may have members that exhibit properties desirable in a photodynamic therapy photosensitizer. These three photosensitizers are SiPc(OSi({dollar}rm CHsb3)sb2(CHsb2)sb3{dollar}- {dollar}rm Nsp+(CHsb3)sb2{dollar}(n-{dollar}rm Csb{lcub}12{rcub}Hsb{lcub}25{rcub})rbracksb2{dollar}2I{dollar}sp{lcub}-{rcub}{dollar}, Pc 14; HOSiPcOSi({dollar}rm CHsb3)sb2(CHsb2)sb2Csb5Hsb4Nsp+CHsb3{dollar} I{dollar}sp-{dollar}, Pc 37; AlPcOSi({dollar}rm CHsb3)sb2(CHsb2)sb3Nsp+(CHsb3)sb2{dollar}(n-{dollar}rm Csb{lcub}12{rcub}Hsb{lcub}25{rcub}) Isp-{dollar}, Pc 21. The results of photobiological studies of the 21 photosensitizers done by collaborators show that one of these photosensitizers, Pc 28, exhibits under specific conditions greater uptake with Chinese hamster V79 cells than Pc 4 and greater photocytotoxicity towards them. The results of these studies and photophysical studies done by other collaborators lead to tentative conclusions about relationships that exist between the structures of these photosensitizers and their photobiological activities.
