In this paper,the fermentation condition of Schizochytrium sp.was optimized,and the yield of docosahexaenoic acid(DHA)was determined by different methods of fluidization and the effect of temperature on the yield of DHA.Crack pot of microalgae oil was applied to study the protective effects of the pathophysiological changes in diabetes mice,and following analyzed the improvement of the body's inflammatory response and renal injury in these diabetes mice.Furthermore,the molecular mechanism regulating the renal injury was further investigated,which will provide the theory basis for the prevention and treatment of renal injury in the diabetes mice through the future use of fatty acids in diet.1.In order to obtain a stable yield of DHA,the optimal fermentation conditions for DHA production by chlamydomonas schizophyllum were established in 5L fermentation tank.After optimization,the conditions were as follows: the acceleration of glucose flow in the middle fermentation period(42?88 h)was 1.5g /L/h using variable flow feeding strategy;Late fermentation(88?120 h)glucose,flow acceleration of 1.0 g/L/h)and two phase temperature control strategy(0?40 h,temperature 30?;40 h until the end of the fermentation temperature 25?),under the condition of the DHA production reached 15.51 g/L.2.This paper verified that feeding diabetic mice with DHA rich algal oil could improve the chronic inflammatory state of the body.After feeding intervention,insulin sensitivity increased and insulin resistance decreased.After the intervention,the total cholesterol,triglyceride and low-density lipoprotein of diabetic mice decreased significantly,while the high-density lipoprotein increased significantly.Algal oil rich in DHA played a strong role in the regulation of lipid metabolism and the improvement of antioxidant capacity of the body,and protected the kidney injury of diabetic mice.3.Further investigation demonstrated and proved that DHA-rich algae oil could decreased the oxidative stress response of diabetic mice and inhibit the activation of NLRP3 inflammatory corpuscles and the expression of IL-1 beta in the renal cortex of diabetic mice with oxidative stress and inflammation,thus delaying the inflammatory reaction and pathological damage.4.Finally,the present paper conducted the gene expression clustering and differential gene analysis of renal cortex in different groups of diabetic mice and found that the gene expression pattern of diabetic mice with feeding intervention was similar to that of normal mice.The differentially expressed genes were significantly enriched in major signal pathways such as NF-?B,p53 and TNFs.It was found that Caspase1\NLRP3 protein and families related to the occurrence of inflammation such as IL and TNF were involved in the process of renal inflammation,suggesting which may play an important role in inhibiting inflammation and protecting the kidney.
