| This study proposes to clarify the characterization of the factors towards the effective component formation and quality regulation of Cistanche tubulosa.Experiments and analysis were done in production base in Yutian Xinjiang province and Cangzhou Hebei province.The effects of the living environment,germplasm resources,growth and development on the formation of medicinal quality of C.tubulosa were studied.A new technology for the production of high-phenylethanoid glycosides and high-oligosaccharides was established.In addition,a new method for rapid and non-destructive detection of the product composition of C.tubulosa was established,the main results are as follows:1)The results showed that the contents of effective components of different germplasms of C.tubulosa were different.The highest content of echinacoside and verbascoside was in the purple scale C.tubulosa with the content of 441.40mg/g,and the content of lowest dense fruit varity only 231.8mg/g.The highest level of mannitol was 37.67mg/g,and the purple flower germplasm was the lowest,which was 18.06mg/g.2)The content of main medicinal components showed different change law,the content of echinacoside and verbascoside were decreased with the growth and development of C.tubulosa,and the content of mannitol increased first and then decreased.3)The highest echinacea and verbascoside contents appeared on the mild saline and alkaline land,mannitol contents increased with soil salinity increases.The contents of echinacea and verbascoside under the condition of 7.0<pH<8.5 were higher than the conditions of pH>8.5 significantly.There was no influence of the solil pH on the content of mannitol in this study.4)The β-glucosidase was able to decompose the PeGs of the C.tubulosa in the process of natural drying.The contents of echinacoside,cistardide A and verbascoside of 5mg β-glucosidease treament were 92.9%,84.8%and 82.7%of control treatment,respectively.Compared with air drying,the contents of echinacoside and verbascoside were increased by the treaments of stoving,steam treatment,hot water blasting and microwave drying.The optimization processing technology was steam treatment with 7 minute.5)In summary,the optimization of the high-PeGs products are to select the purple scales purple flower C.tubulosa germplasms,select the mild saline and alkaline land(7.0<pH<8.5,0.2%<soil salinity<0.4%)and the fresh weight of C.tubulosa less than 250g.Fresh C.tubulosa are cut into slices with 1 cm and steam treatment with 7min.The optimization of high-manntilo products are to select the black scales black purple flower germplasm resources,choose pH below 8.5 and the fresh weight more than 250g.Fresh C.tubulosa are cut into slices with I cm and steam treatment with 7min.6)In this study,we established a new method of non-destructive and rapid determination of the contents of C.tubulosa.Near infrared spectroscopy can be used to predict the contents of echinacoside,verbascoside,mannitol,fructose,sucrose and glucose in the fresh and powder sample of C.tubulosa. |
