| Cistanche deserticola Y.C.Ma is a perennial parasitical plant of Orobanchaceae Cistanche. its host are Haloxylon ammodendron (C.C.May) Bge) and Haloxylon Persium in northwest area of China. This experiment selects the artificial cultivation Cistanche deserticola Y.C.Ma as tested materials, the content of phenylethanoid glycosides, determination of echinacoside and acteoside, with total yield as the index, the extraction process of Herba cistanches preliminary judgement. The main conclusions were as follows:1Determination Methods of Cistanche and extract PeGs components of HPLC: Discovery C18column (25cm×4.6mm,5μm); mobile phase ratio and keep time for methanol:0.1%formic acid (26.5:73.5) for17min, compared to methanol:flow change within3min formic acid (29.5:70.5), must maintain15min; flow rate:1mL·min-1; detection wavelength:330nm; column temperature:32℃; sample size:10μ L; simultaneous determination of two active ingredients, the baseline was more stable, less interference, good degree of separation, the study on the method found the established HPLC method for the determination was reliable.2The research of different drying temperature of fresh Cistanche affect effective component content:Drying temperature of85℃and105℃were better, which could keep two kinds of effective component content higher.3Study on the preservation time of oil Cistanche and non-oil Cistanche vacuum fresh-keeping, two main effective component content was stable within90d, when more than120d. decreas of the content of effective components was obvious. The content of effective components in non-oil Cistanche in relatively stable within30d. when more than60d, the content of effective components decreased obviously.4The yield of Cistanche main effective components of echinacoside and acteoside as investigation index. Preliminary exploration and extraction of Cistanche two components were determined:The ideal of20~80mesh effect crushing herbs; ethanol was used as extraction solvent; ethanol concentration60%~70%; the ratio of solid to liquid1:10; extraction times of2; every time3~4h. The effective component yield were higher in this condition. |
