In the present study, we aim to explore the relationship between vascular invasion and tumor metastasis by dual immunohistochemical staining of the sections of tongue squamous cell carcinomas patients, and to assess the association of eNOS, ICAM-1, E-selectin polymorphism with oral cancers. This study consists of two parts:(1) the difference of the penetration of cancer cells into blood vessels and lymphatic vessels, and the relationship between vascular invasion and tumor metastasis; (2) the Glu298Asp and 27 bp VNTR polymorphism of eNOS gene, K496E polymorphism of ICAM-1, and S128R polymorphism of E-selectin in oral cancer.Part One:Vascular Invasion In Tongue Cancer And Tumor MetastasisObjective To explore the difference of the penetration of cancer cells into blood vessels and lymphatic vessels, the relationship between vascular invasion and tumor metastasis, to detect the expression of E-selectin and ICAM-1 in invaded vasculature, and tried to provide a more accurate proof for choosing the proper treatment methods of squamous cell carcinomas of the tongue based on vascular invasion of cancer cells. Methods The wax blocks were collected from 106 patients who suffered from SCC of the tongue. The sections were stained with dual immunohistochemical staining to label the tumor cells and endothelial cells. The expression of E-selectin and ICAM-1 in invaded vasculature was detected by immunohistochemical methods. Data were analyzed by SPSS 13.0 Statistical Software. Results Vascular invasion was detected in 44 cases. Among these cases, blood vessels invasion was present in 39 cases; lymphatic vessels invasion was found in 2 cases; and both vessels invasion was observed in 3 cases. E-selectin and ICAM-1 strongly expressed in invaded blood vessels in those cases with cervical lymph node metastasis while weakly or negatively expressed in those cases without lymph node metastasis. All lymphatic vessels didn't express E-selectin and ICAM-1. Conclusion The blood vessels invasion but not lymphatic vessels invasion was positively associated with cervical lymph node metastasis. The difference of the penetration of cancer cells into the vasculature may be related to the expression of E-selectin and ICAM-1. Part Two:eNOS, ICAM-1, E-selectin Polymorphism in Oral CancerObjective To investigate whether the Glu298Asp and 27 bp VNTR polymorphism of eNOS gene, K469E polymorphism of ICAM-1, S128R polymorphism of E-selectin were associated with oncogenesis and development of oral cancer. Methods Blood samples from 112 oral caner patients and 98 control subjects were recruited to the study. The 27 bpVNTR and Glu298Asp polymorphisms of the eNOS gene, K469E polymorphism of ICAM-1 gene, S128R polymorphism of E-selectin gene were detected by PCR- FELP. We compared genotype frequencies and allele gene frequencies between oral cancer group and control group. Results The data showed that the 27 bpVNTR4a/4a,4a/4b genotypes and Glu298AspGG, GT genotypes of the eNOS gene, K469E EE genotypes of ICAM-1 gene, S128R AC genotypes of E-selectin gene were more common in cancer cases than control(P<0.05). The allele gene of 4a(OR=15.321; 95%CI:9.273-25.315), T(OR=2.816; 95%CI:1.600-4.959), E(OR=2.076; 95%CI:1.400-3.077), C(OR=2.795; 95%CI:1.638-4.771) increase the risk of oral cancer,and the allele gene of C(OR=1.875; 95%CI:1.080?3.255) increased the risk of oral cancer metastasis. Conclusion The Glu Asp and 27 bp VNTR polymorphism of eNOS gene, K469E polymorphism of ICAM-1, S128R polymorphism of E-selectin were associated with oral cancer.
