Study To The Effect Of IL-10 On ICAM-1 After Spinal Cord Injury

Objective To evaluate effect of IL-10 on intercellular adhensionmolecular - 1 (ICAM-1) expressed on the surface of spinal cord blood vessels endothelium cells after secondary spinal cord injury.Methods Immunohistochemistry was used to observe the expressionchange of ICAM-1 as result of IL-10 and MP 1,4,12,24,48,72,120 hour respectively after SCI.105 healthy big rats were choosed and their heavy varied from 280 gram to 310 gram. Big rats were divided into five experiment group randomly: controlled group, pure handled group, IL-10 handled group, MP handled group, MP and IL-10 handled group. The change of ICAM-1 in each group were observed after 1,4,12,24,48,72,120 hour. Each time point have there big rats. MP handled group infused MP 30 milligram each kilogram into vein. IL-10 handled group infused reorganized rat IL-10 15 ug each kilogram into belly cavity . Only vertebral plate were cut , but spinal cord were not hurt in controlled group. Not only vertebral plate were cut , but also spinal cord were hurt in pure handled group. But they were not handled by IL-10 or MP. Big rat spinal cord injured model was established with ALLEN bump equipment. ICAM-1 in the injured spinal cord was applied with Immunohistochemistry methods to dye. Slices were counted with multipled 400 light lens. Different district were choosed randomly. Two districts were not repeated. Four districts were counted in the gray matter and Six districts were counted in the white matter.Results ICAM-1 expression was not observed 1 hour , began to appear inthe part of injury 4 hour, reached its peak 24 -48 hour after SCI, after that i t decreased gradually . ICAM-1 could be still observed 120 hour after SCI . ICAM-1 express mainly in endothelium cell .ICAM-1 concentrate in the gray matter nearby central tube . Endothelium cell of blood vessels in the artery and vein all express. ICAM-1 was not asymmetric in the spinal cord endothelium cell. The gray matter is stronger than the white matter .Centraldistrict was more obvious than the edge dropsy district . 1L-10 and MP obviously inhibited the expression of ICAM-1 . IL-10 inhibited ICAM-1 obviously after 4,12,24,48,72 hour when spinal cord were injured. But it have not function after 120 hour. IL-10 inhibited ICAM-1 more obviously in the gray matter than in the white matter. IL-10 inhibited ICAM-1 more obviously in the central district than in the edge dropsy district. MP have more stronger effect than IL-10, which became more obviously when IL-10 and ICAM-1 were combined.Conclusions ICAM-1 play a role in the early pathological stage ofinjury. It is one of the important factors that result in secondary spinal cord injury. Although IL-10 and MP inhibit ICAM-1 in the early stage of SCI, both of them have no effect on ICAM-1 in the later period. Because ICAM-1 can mediate the adhension between white cell and endothelium cell ,which speed up the infiltration of white cell in the area of injury spinal cord , IL-10 and MP blocking the expression of ICAM-1 in the early stage of SCI are useful to alleviate the extent of injury after SCI.
