ObjectiveOral and maxillofacial bone tissue is an important supporting structure and the reconstruction of the defect is very important.Tissue repair often requires scaffold materials to support cells and delivery factors.As a new type of inorganic material,graphene materials need a lot of exploration on how to properly apply it to the loading of protein factors and modification of composite materials.Therefore,this study aims to improve the material system by constructing gelatin reduced graphene oxide(GOG)and photocrosslinked hydrogel composites with GOG loaded with drug,which provided new available materials for bone tissue repair.Materials and MethodsTo improve the material system,the gelatin reduced graphene oxide(GOG)was firstly obtained by using gelatin reductivity.Its physicochemical properties and drug loading properties were characterized and its biological activities(endocytosis uptake and pharmacodynamic enhancement)were also evaluated.Second,GOG is used as filler to build photocrosslinked hydrogel composites(PH/GOG).By characterization of the physical and chemical properties,the optimal content of GOG in the composites is determined.Furthermore,we constructed the photocrosslinked hydrogel composites with GOG loaded with drug,and a series of experimental tests in vivo and in vitro were conducted to evaluate its contribution to the bone tissue repair in terms of promoting the forming of the bone and blood vessels.Meanwhile,we also explored its application prospect as carrier material for the delivery of the drug.ResultsThrough the study on GOG,it was shown that GOG could rapidly enter cells and widely distribute in the nucleus and cytoplasm,and it also had good cellular compatibility and didn't affect the normal vital activity.The study about its drug loading capacity and synergistic effect showed that GOG could also enhance the efficacy of the drug.Studies on the composites showed that the photocrosslinked hydrogel composite with appropriate proportion of GOG loaded with drugs was beneficial to the osteogenic and angiogenic differentiation of the cells,which promoted the rapid and effective repair of defects.ConclusionIn conclusion,GOG had good performance in drug delivery and the photocrosslinked hydrogel composites with GOG loaded with drugs could promote osteogenic and angiogenic differentiation of the cells.The studies about the modification of materials provided a new potential material system for bone tissue repair.
