Objective:To observe the protective effect of salvianolic acid B preconditioning on intestinal ischemia-reperfusion rats,and to study its possible mechanism.Methods:This experiment set the negative control group and model group,Dan phenolic acid B dosage group has also set up the corresponding control group and model group,each group of 12 random equipped with male SD rats,through the clip the superior mesenteric artery,make the intestinal ischemia,maintain 60 min,24 h after reperfusion experiments needed for intestinal ischemia reperfusion model is obtained.The contents of CAT,SOD,GSH and MDA in the serum of each group of rats were measured.The contents of CAT,SOD,GSH,MDA,tnf-alpha,il-1 beta and il-6 in the ileum of each group of rats were measured.The activity of medullary peroxidase(MPO)and the level of nf-kappa B p65 were detected.Determination of intestinal mucosal permeability using FITC combined with dextran.The ileum tissue of 1cm was taken for HE staining to observe the pathological changes of the corresponding section,and Chiu's score was performed.The correlation between the contents of tnf-alpha,il-1 beta and il-6 and the expression of nf-kappa B p65 was analyzed.The correlation between the levels of tnf-alpha,il-1 beta,il-6,nf-kappa B p65 and Chiu's score was analyzed.In addition,rt-pcr and Western Blot were used to determine the m RNA expression and protein level of pi3kp85 a and p-akt(ser473)in the ileal tissues of rats in each group.Results:1.The serum MDA level of the model group was significantly higher than that of the negative control group,while the antioxidant SOD,CAT and GSH levels were significantly lower(P<0.01).Compared with the model group,the content of MDA in serum of salicylic acid B+ model group was significantly decreased,while the content of SOD,CAT and GSH were increased,the difference was statistically significant(P<0.01).2.Compared with the negative control group,the content of MDA in the ileum of the model group was significantly increased,while the antioxidant SOD,CAT and GSH all showed a significant decrease trend,with statistically significant difference(P<0.01).Compared with the model group,the level of MDA in the ileum of rats in the salicylic acid B+ model group decreased significantly,while the antioxidant SOD,CAT and GSH increased significantly(P<0.01).3.Compared with the negative control group,the contents of tnf-alpha,il-1 beta and il-6 in the ileal tissues of the model group were significantly increased(P<0.01).Compared with the model group,salvianolic acid content in B+ model group was significantly lower than that in the model group,and the difference was statistically significant(P<0.01).4.Compared with the negative control group,MPO and nf-kappa B p65 levels of ileum homogenate in the model group were significantly increased,and the difference was statistically significant(P<0.01).Compared with the model group,the MPO level and nf-kappa B p65 in the model group were significantly decreased(P<0.01).5.Compared with the negative control group,the content of fitc-glucan in the serum of the model group was significantly increased,and the mucosal permeability was increased.Compared with the model group,the content of fitc-glucan in plasma of salvianolic acid B+ model group was decreased,and intestinal mucosal permeability was decreased(P<0.01).6.Compared with the negative control group,the intestinal mucosa of rats in the model group was seriously damaged,capillaries were ruptured,resulting in impaired glandular function,neutrophil infiltration was found,and Chiu's score was significantly increased(P<0.01).Compared with the model group,the intestinal mucosa of rats in the salvianolic acid B+ model group was slightly damaged,and the Chiu's score was decreased(P<0.01).7.In the process of intestinal ischemia and reperfusion injury in rats,the correlation between il-1 beta,il-6,tnf-alpha and nf-kappa B p65 was analyzed and found to be significantly positive,with the corresponding correlation coefficients0.932,0.949 and 0.937,respectively(P<0.01).The contents of il-1 beta,il-6,tnf-alpha and nf-kappa B p65 in the ileum of rats after intestinal ischemia and reperfusion were significantly positively correlated with the Chui's score of ileum,with correlation coefficients of 0.901,0.884,0.873 and 0.898,respectively(P<0.01).8.Compared with the negative control group,the m RNA expressions of pi3kp85 a and p-akt(ser473)in the ileal tissues of the model group were significantly decreased(p <0.01).The m RNA expressions of pi3kp85 a and p-akt(ser473)in ileum in the B+ model group were significantly higher than those in the model group(p<0.01).9.Compared with the negative control group,the protein expression of pi3kp85 a and p-akt(ser473)in the ileal tissues of the model group was significantly decreased(p <0.01).Compared with the model group,the protein levels of pi3kp85 a and p-akt(ser473)in the ileum of the B+ model group were significantly increased(p <0.01).Conclusions:1.Salvianolic acid B has a certain protective effect on the rat model of intestinal ischemia reperfusion injury,while the oxidation resistance,improvement of intestinal mucosal permeability and inhibition of inflammatory response are all affected by its action mechanism.2.Salvianolic acid B exerts its limiting effect on the expression of IL-1 ?,IL-6and TNF-? by regulating the signal pathway of NF-? B.3.Salvianolic acid B may alleviate and improve intestinal ischemia-reperfusion injury by regulating PI3K/Akt signaling pathway.
