Atherosclerosis(AS)is one of the major pathogenic factors and the cause of death throughout the world,characterized by slow onset,long disease cycle and multiple risk factors.Multiplication of vascular smooth muscle cells(VSMC)can greatly contribute to the development of atherosclerosis.There is still lot of controversy on which transition proliferation inhibition of VSMC has benefit to the treatment of AS.GXⅡ(GXⅡ)is a famous traditional Chinese medicine formula,little research results have been reported at this point concerning the anti-atherosclerosis effect of GXⅡ.In this study,the anti-atherosclerosis mechanism of Ferulic acid(FA),the effective component from GXⅡ has been described systematically via the animal models of AS and VSMC in vitro experiments.The research structure is as follows:to test the compounds of GXⅡ formula by UPLC,then to detect the absorbed bioactive compounds(ABC)in serum via UPLC-MS/MS to confirm that FA is one of the main ABCs in blood.To stable the AS rodent model with ApoE-/-mice feed on high fat diet.Observing the effect of GXⅡformula,FA,and Simvastatin anti-atherosclerosis respectively,the results show that all three drug therapy groups can lower the blood lipids level and reduce atherosclerotic plaque area when compared with AS model group.FA was found to inhibit platelet-derived growth factor(PDGF)promotion of VSMC migration and propagation in in vitro cellular assays,and make the VSMC cycle stay in G1 phase by promoting VSMC nitric oxide generated and increasing expression of P21,eventually lead to the stagnation of the cell cycle related cycle protein,give play to the role of inhibition of cell proliferation.Overall,this study has shown that FA,as the main ABC of GXⅡ play some part role in anti-atherosclerosis through the NO/P21 pathway to inhibit the VSMC proliferation.In addition,the result also support that FA inhibit VSMC proliferation in early intervention is benefit to treatment of the AS.The further study on whether to inhibit VSMC proliferation does some well in the atherosclerosis plague stable is still needed.the result of this research indicate that FA may be a potential drug to treat the proliferation of blood vessels.
