| Purpose:Phlegm syndrome and blood stasis syndrome are the common TCM Syndromes of non-small cell lung cancer(NSCLC).In this study,Erchen decoction was used to intervene NSCLC to clarify the intervention effect of phlegm and blood stasis syndrome,as well as the influence of Erchen decoction on tumor angiogenesis related indicators and tumor invasion and metastasis of NSCLC.1.Theoretical study on the treatment of non-small cell lung cancer with traditional Chinese MedicineObjective:To study the relationship between Yiqi phlegm therapy for non-small cell lung cancer and the relationship between blood stasis syndrome and angiogenesis and invasion and metastasis of lung cancer.Methods:The origin,relationship and key terms of anti-tumor of traditional Chinese medicine were combed,and the theoretical research and experimental results of Yiqi Huatan method in the treatment of non-small cell lung cancer were analyzed and discussed from the perspective of Yiqi Huatan method,and the blood stasis syndrome and tumor angiogenesis The relationship between invasion and metastasis explored the theoretical connotation of Erchen decoction in the treatment of non-small cell lung cancer.Results:The pathogenesis and development of non-small cell lung cancer in the middle and late stage are closely related to the pathological products such as phlegm and blood stasis.Therefore,there are many patients with mixed syndrome of qi deficiency,phlegm and blood stasis in clinic.On the treatment,the two Chen Decoction with a flat and simple and delicate drug is selected as the foundation,and then according to the proportion of the positive and weak evil and deficiency of the patients,The drug with the advantages of Invigorating Qi and nourishing Yin,tonifying positive Qi,activating blood circulation and eliminating blood stasis,and eliminating cancer by Pingdu can we flexibly respond to the phlegm stasis and get back and forth in the clinical medication.2.The analysis of the network pharmacology of Erchen decoction in the treatment of nonsmall cell lung cancerObjective:To predict the possible mechanism of erhangtang in the treatment of non-small cell lung cancer by using the method of network pharmacology,so as to explore the potential therapeutic targets of ERC,and provide theoretical basis for experimental research.Methods:1.The relevant target points of four basic drugs in erhangtang can be retrieved by logging in the pharmacology data of traditional Chinese medicine system,tcmsp and tcmid respectively;2.to search the disease related targets of genecards,OMIM,CTD,KEGG and other databases by NSCLC and non small cell lung cancer;3.map the predicted disease-related targets and drug targets to construct a protein interaction network of protein protein(PPI),and calculate the topological structure,so as to get the core target of drug diseases and display them;4.the selected drug disease target was introduced into Davis analysis and KEGG pathway was enriched.The molecular mechanism of Erchen decoction in the treatment of NSCLC was discussed.Results:1.22821 NSCLC disease related targets were predicted.2.63 drug disease core targets were selected by setting relevant parameters.3.55 signal pathways related to the treatment of NSCLC were obtained by the analysis of pathway enrichment.The top 10 pathways included TNF signal pathway,pi3k/akt signal pathway,HIF-1 signal pathway and AMPK signal pathway.4.Erhangtang can inhibit angiogenesis,immune regulation,tumor invasion and metastasis through many ways,among which pi3k/akt signal pathway and HIF-1 signal pathway can be used as important pathway for treatment of NSCLC.3.Experimental study on the antitumor effect and mechanism of Erchen decoction on Lewis miceObjective:To observe the effect and mechanism of the expression of PI3K,Akt,VEGF,VEGFR-2,Gli-1,SHH,SMO,Ptch-1 and so on in Lewis lung cancer mice.Methods:40 C57BL/6 mice were established by subcutaneous inoculation of Lewis lung cancer cells.After the model was successfully established,the model was divided into model group,traditional Chinese medicine group,combined group and cisplatin group,each group was 10.The traditional Chinese medicine group and the combined group began to give the gastric administration on the 7th day after the model was made,twice a day,for 14 days.The model group was given equal amount of physiological saline.Cisplatin group was given 200mg/kg cisplatin solution 200ul/20g for intraperitoneal injection,once a day,for 3 days,and 0.4ml/20g of normal saline for stomach twice a day for 14 days.Observe the general situation of mice and tumor growth.14 days later,the tumor tissue was removed and blood was taken from the eyes.The blood routine level was detected by routine biochemical method;The expression levels of PI3K,Akt,CD34,VEGF,VEGFR-2,SMO,Gli-1,Ptch-1 and Shh were detected by immunohistochemistry,Westernblot and RT-PCR.Results:1)Both the Chinese medicine group and cisplatin group can improve the tumor growth and tumor inhibition rate of mice.The tumor inhibition rate of the combined group is higher than that of other groups,but there is no significant difference with cisplatin group(P<0.05)。2)WBC and RBC of the combined group,mice were lower than those in model group and traditional Chinese medicine group(P<0.05),and hemoglobin count of the combined group was higher than that of cisplatin group(P<0.05).PLT of cisplatin group and combined group was significantly higher than that of other two groups(P<0.05).3)the expression of MVD,CD34 in the tumor tissue of the combined group was significantly lower than that of other groups,and decreased with the increase of concentration in a certain concentration range,the difference was statistically significant(P<0.05).4)the protein expression of PI3K,Akt,CD34,VEGF,VEGFR-2,SMO,Gli-1,Ptch-1 and Shh in tumor tissues of mice was inhibited by traditional Chinese medicine group and the combination group(P<0.05).Conclusion:the mechanism of erchengtang on NSCLC is multi-target and multiple cell pathways interact with each other,which is closely related to the angiogenesis,inhibition of tumor invasion and migration.Compared with the simple chemotherapy,the combination of Ercheng Decoction and chemotherapy has a certain effect on NSCLC patients,which can improve the quality of life and reduce the side effects of chemotherapy;The combination of Erchen decoction and chemotherapy can effectively improve the quality of life and reduce the side effects of chemotherapy.Erchen decoction can significantly inhibit the proliferation and cloning of Lewis mice.It can regulate the angiogenesis related VEGF/VEGFR-2 protein family through pi3k/akt pathway.Compared with the control group and the traditional Chinese medicine group,the protein levels of PI3K,Akt,VEGF,VEGFR-2 and other proteins have significantly decreased,After treatment with Erhang decoction,the levels of Shh,SMO,PTCH and Gli-1 decreased with the treatment of Lewis mice;It suggested that Erchen decoction had the ability to inhibit the invasion and migration of tumor cells. |
