| Objective: Application of high performance liquid chromatography and high resolution mass spectrometry to explore the quality evaluation methods of Traditional Chinese Medicine derived from the genus Citrus and dispensing granules.Methods: Literature analysis of citrus flavonoids by bibliometric method;application of high performance liquid-phase fingerprinting technique combined with machine learning classification algorithm to classify citrus Traditional Chinese Medicines.Automatic identification of chemical constituents of Daidai Zhishi,Daidai Zhiqiao,Suancheng Zhishi and Suancheng Zhiqiao using UNIFI platform based on self-built database.Application of high performance liquid chromatography to establish quality standards for dispensing granules of Juye.Results: The results of the analysis of the bibliometric method showed that the number of articles on citrus flavonoid research is still increasing,and most of the articles were published in journals in the field of food and pharmacology.The biological activities of citrus flavonoids are extensive,mainly involving oxidative stress,apoptosis and metabolism,and the related diseases are cancer,neurodegenerative diseases and metabolic diseases,and hesperidin,naringenin and nobiletin are the ones that appear more frequently.A high performance liquid-phase fingerprinting method based on citrus flavonoids was established to test 367 batches of citrus TCM samples,and the classification accuracy of eight classification models on the citrus TCM fingerprinting dataset was compared,and it was finally determined that Decision Tree Classifier and Extra Tree Classifier were more effective in classifying the dataset,and the optimization of model hyper parameters,the classification accuracy could be stably maintained at 98-100%.A database including 754 chemical components of citrus herbal medicines was established,and the UNIFI platform was applied to identify 188 components based on the high-resolution mass spectrometry data and compound database for the chemical composition identification of Suancheng Zhiqiao,Suancheng Zhishi,Daidai Zhiqiao,and Daidai Zhishi.The fingerprint profiles and content determination assays of dispensing granules of Juye were established.Conclusion: Citrus flavonoids are still a hot research topic.The high performance liquid-phase fingerprinting method of citrus flavonoids can detect the eight flavonoid components contained in citrus TCM,and Decision Tree Classifier and Extra Tree Classifier can realize the classification of citrus TCM with the accuracy of more than 98%.The UNIFI platform was applied to identify 188 chemical components in Suancheng Zhiqiao,Suancheng Zhishi,Daidai Zhiqiao,and Daidai Zhishi,and there were differences in the chemical components of Daidai Zhishi and Suancheng Zhishi from different origins.The results of the methodological investigation showed that the established method for the characterization mapping and content determination of dispensing granules of Juye was accurate and reliable. |
