Myxomycetes are a special group with characteristics both of animal and eumycetes. They widely inhabit in the world, surviving in the environments of suitable temperature and moisture, where plants and fungus remnants live. Myxomycetes has plasmodium (ameba) stage like animals and spore stage like fungi in its life cycle. Therefore, its taxonomic position is still unclear. The taxonomic studies on Myxomycetes are needed by new method and technique.In order to investigate the biological and chemical proofs of systematic taxonomy of Trichiales myxomycetes, studies on ontogeny, soluble protein and lipo-soluble chemical components of main species of Trichiales myxomycetes were carried out by using the method of moist chamber culture, media culture, SDS- polyaccrylamide gel electrophoresis, GC/MS, the life cycle of main species was observed, the relationships between main species were clarified, the culture media and condition were obtained, and 4 compounds from myxomycetes were found for first time. This research will results in the further study on medicinal fungi resource.It is helpful to understand the relationship on ontogeny between the species of Myxomycetes by laboratory culture. The taxonomic knowledge can be gotten by comparing the different stages of lifecycle between the species. More than 16 species including 5 Trichiales were obtained by moister chamber culture. Different chamber culture, temperature, moisture, illumination, and the effect of chamber cultures were discussed. The observation over the unit growth process of myxomycetes by moister chamber culture was made to prove that moister chamber culture was not restricted by seasons. The findings indicate that moister chamber culture is a satisfactory approach to obtain myxomycetes in laboratory condition, and make up for the insufficiency of field collection. The culture method was modified to fit one species, Trichia favoginea, in which it is difficult to succeed in media culture. The entire process was studied on it from spore to spore in solid media culture.The characteristics of ontogeny in different stages were observed.The optimal culture condition of Trichiales myxomycetes is: the spores are cultured in corn media with extract solution from dead wood, then transfer the plasmodia to oat media with pH8, at 281, adding water per 2 days and giving scattered light for 0.5h per day. The modified culture method will provide the materials of myxomycetes.As we known, the characteristics is the result of both genetics and environment. The molecular evidence can give us the knowledge of phylogeny and the relationship between species. Polyaccrylamide gel electrophoresis was used in taxonomy study. The results shows that the bandstyle of each species in SDS - PAGE reflect has speciality, it can reflect the relationship between the species. The band shows that there are few same sub-proteins among the order of Trichiales. The same sub-proteins are fewer among genea than within a genus. And it is same within a species. Protein is transfered by RNA coded by DNA and can reflect genetics. This results shows that Polyaccrylamide gel electrophoresis can be a good proof on molecular level in taxonomy of myxomycetes.The lipo-soluble compounds from main species of Trichiales were isolated and analyzed by GC/MS. Four compounds from myxomycetes were found for first time. They are: Phosphoric acid tributyl ester, Butanediotic acid, methyl-, bis (1-methyl propyl) ester, 1,2-Benzenedica rboxylic acid dibutyl, and Heptadecane. The same compounds were found both in plasmodia and fruiting body from same species, but the components from different species are different. It shows that the components of myxomycetes speciliality within a species. The chemical compounds can reflect the relationship between species and can be a proof in the taxonomy of Myxomycetes. The compound Phosphoric acid ester can inhibit the activity of phosphorase and can kill the cells of some cancer according to the reporting.
