Objective:Study the mechanism of radiation injury by # -ray radiating NIH3T3 and SHG44 cell lines with electron linear accelerator.At the same time.stuty the radioprotection of Taurine by adding different concentrations Taurine to the cell nutrient before # radiation with 15Gy.Methods:(l)Observe the change of cell cycle by flow cytometer.(2)Detect the cell proliferation conditions with the MTT method.(3)Mesure the expression MMPl -, MMP2, ppGalNAcT2 mRNA by reverse transcription polymei ase chain reaction(RT-PCR). Results:(l)The NIH3T3 and SHG44 cell lines all shewed the G2/M phase arrest after 12h of radiation.The proliferation of SHG44 cells was inhibited.Both were related to the dosage of irradiationThe delay of G2/M phase alleviated when adding Tau, which became more obvious with the increase of Tau's dosage. (2) After # radiation,the expressions of MMP2 mRNA in both cell lines were increased and the expression of MMPl mRNA in NTH3T3 cell lines;meanwhile,a decrease of ppGalNAcT2 expression in both cell lines was was observed.When Tau was added to the cell culture,the expression of ppGalNAcT2 mRNA in both cell lines was increased.but the expressions of MMPl , MMP2 in NIH3T3cell lines and the expression of MMP2 in SHG44 cell lines were inhibited,whichConclusions:(l)high enegy electronic can not only injury normal cells through inducing cell denature,but also has influence on tumor cells probably through increasing theie ability of invasion and migration.(2)Tau decreases the dissociation rate of normal cells and weakens the ability of invasion and migration in tumor cells by the down-regulation of MMPs mRNA expressions and the up-regulation of ppGalNAclT2 mRNA expression.Tau improves the repairmen! ability of normal cells and inliibits the proliferation of tumor cells by extension Gl phase in cell cycle.
