Polyamines (spermine, spermidine and putrescine) are simple linear aliphatic compounds presenting in almost every cell, which play important biological roles in plant growth and development and have been implicated in many physiological functions, including cell division and differentiation, nucleic acids replication, transcription, translation, synthesis of proteins and membrane stability. More attention has been paid to explore analytical method of polyamines for high selectivity, sensitivity, repeatability and minimum running cost.Resent years, the chemiluminescence (CL) technique has become an essential tool in routine trace analysis because of its high sensitivity and simple instrument. In this thesis, it is firstly discovered that an unsaturated complex of Cu( II) and polyamines has a stronger catalytic effect on luminol-H2O2 chemiluminescence reaction. Based on this mechanism, a new liquid phase analysis method is established by using weak chemiluminescence analyzer for detection of spermine. At the optimum conditions, the detection limit is 4.0x10-9 mol/L, the relative standard deviation (RSD) for 1.0×10-7 mol/L spermine is 5.6% (n = 5) and the linear range locate in 1.0×10-5 mol/L 1.0×10-7mol/L. Compared with the method of derivatization and fluorescence detection, this method is characterized by more sensitivity, simple operation and wider linear range.A solid Cu(OH)-2 column chemiluminescence method combined with flow injection technology (FIA-CL) was also established based on the catalytic effect of polyamine-Cu( II) complex on luminol-H2O2 chemiluminescence reaction, in which the unsaturated complex of Cu( II) and polyamines was formed when polyamines flow through the solid Cu(OH)2 column. Compared with the stabilization enzymatic chemiluminescence detection system, this method has the advantage of simple instrumentation, time-saving and more sensitivity.
