The microstructure of normal retina and experimental retina in the Carassius auratus were observed and described in detail. The experimental retina was the retina of Carassius auratus after the optic never damage. Our attention was paid to the optic system in Carassius auratus. We made quantitative analysis of thickness of the normal retina, every normal retinal layers, the experimental retinal and every retinal layers with sections stained by H.E. Meanwhile,the expression pattern of GFAP in the retinal-tectal system of Carassius auratus during the regeneration of optic nerve was studied, using immunohistochenistry. The results were following:(1)The retina of Carassius auratus was similar with the retina of most vertebrates. And there were ten layers in the retina, but thickness of ONL, INL and IPL were equal, the character of the retina belonged to the night animals'. (2)The thickness of some retinal layers decreased firstly, then increased and decreased to the level of normal retina in the compare between the normal and the experiment.(3) In the compare between the contrast and the experiment .the result showed that the change of the thickness of PEL and PRL was not distinct, and the changes of the thickness of rest of the retina were distinct, the changes of the thickness of every retinal layers in 5d and 14d after optic nerve damage were highly distinct.(4)The results of the immunohistochenistry told that GFAP expressed in IPL, NF, INL, and OPL of the experimental retinal. In normal retina, GFAP expression was found in NF, ILM, INL, OPL.the rest of normal retina were not found the expression of GFAP.(5)ln the normal tectum, GFAP was highly in SO, and the lower expression of GFAP was found in fewer cell of SFGC, SAC, SGC. In the experimental tectum, GFAP in SM.SFGC, SAC, SGC remained to control level .the expression model of GFAP in SO was increased firstly, and then decreased peaking at 5d, but it was no less than the normal tectum. In the experimental tectum, the expression of GFAP in the right SO was higher than the expression of GFAP in the left SO during 1d-60d after after optic nerve damage. The expression of GFAP in the right SO was familiar with the expression of GFAP in the left SO during 85d after optic nerve damage, but the expression of GFAP in SO in the experimental tectum was higher than that in the normal tectum. The result of GFAP expression in the experimental retina and tectum showed that AStrocyte(muller cell) took part in the optic nerve regeneration and it might play an important role in optic nerve regeneration accidents.
