In this experiment, ultrafiltration and anion exchangeresin were used in the process of purifying the lysozymefrom egg white, including purification,identification anddetermination of enzyme activity.First, by using the elution buffer PBS(Na2HPO4 _NaH2PO4,pH8.5, I0.20mol/kg), the solution was homogenization. A withthe PES membrane(MW=30000Da), 0.35MPa of filtrationpressure, the buffer PBS was used to elute.The part of filtrated was performed by DEAC-celluloseanion exchange resin, under the condition of Trisbuffer(original pH10.0, I=0.1).Then the eluted part was collected, ultrafiltered againand desalinized with the pressure of 0.35MPa.The ultrafiltrate was dried by refrigeration. And thepurity was up to99.5%, enzyme activity was 15500U/mg, thefinal yield is 60%.
