Lysozyme(N-acetylmuramideglyconohydrolase)is one of the bestcharacterized hydrolases.It cleaves β-1,4 linkages of GlcNA chomopolymerand (GlcNAc-MurNAc)n heteropolymer,which causeslysisof the bacteriacon-taining the sepolymers in the cell wall. Lysozyme is considered to be aself-defense enzyme, which is produced in serum, mucus and many organs ofvertebrates. Lysozymes have been classified into three distinct forms by theiraminoacid sequences and tertiary structures, namely, C-type.Althoughlysozyme is found in many organisms, limited information is available onreptile lysozyme. Lysozymes may be important in reptiles,since they livewith opportunistic bacterial infection in the environ-ment.Reptile lysozymewas first purified from the egg white of TrionyxgangeticusCuvier. N-terminalsequencing suggested that this lysozyme was of the C-type.Extractions of lysozymes in egg white are egg white extraction,polyacrylamide adsorption and CM Sepharose FF.通 By CM SepharoseFF,activity is 8 times higher than before. The best temperature of lysozymesis 50℃and pH is 6.0. Cylinder plate method was used to detect the effect onthe fowlling bacteria: Pseudomonas aeruginosa, Staphylocococus aureus,Escherichia coli and different species of resistant Gram-positive. Lysozymehave significant effect in vitro growth inhibition on bacteria.
