This research choose 184 individuals crucians as the study materials including C.auratus TiYu yinji , C.auratus PengZiji, C.auratus Yinji, C.auratus . auratus , C.auratus XiangYunji to analysis genetic diversity and affiliation of populaton using microsatellite DNA .This research adopt seven primers of microsatellite DNA that Crooigmans et al. separated out from the carp {MFW14, MFW16, MFW18 ,MFW24, MFW25, MFW28, MFW29) and one primer (MFW32) according to microsatellite DNA of crucians reported by liu-zhen .There are idiocratic fragments amplified by eight primers from genomic DNA among five breed. The main results as follows :1 We measures 35 alleles from eight microsatellite locus among five breed of crucians ,the bound of alleles number of eight microsatellite locus is 36,and the average alleles number is 4.375, the average alleles number is greater than effective alleles number corresponding in all populations , the bound of average alleles number and effective alleles number of five population is 3 6 and 2.2949~4.9661 respectively.2 Except MFW24(PIC=0.4645) and MFW28(PIC=0.3241), the PIC of other six microsatellite locus reach high polymorphism level in 5 crucians breeds, the bound of PIC number is 0.5389-0.7615, this indicate the crucians breed have prolific genetic diversities . At the same time , we found there has large difference among the average PIC of breed, the average PIC of C.auratus YiYu yinji is the highest (0.6489) and C.auratus PengZiji is the lowest (0.4539), and C.auratus Yinji, C.auratus. auratus, C.auratus XiangYunji is 0.6322, 0.4720,0.6423 seperately.3 The average heterozygosity of five breed is 0.6760, the C.auratus Yinji (0.7269)is the highest , the other three breedsare C.auratus YiYu Yinji(0.7219) ,C.auratus XiangYunji(0.7020) ,C.auratus PengZiji(0.6315) C.auratus auratus is the lowest (0.5978). The average heterozygosity of 8 microsatellite locus is among 0.5388~0.7942, MFW18 is the highest and MFW24 is the lowest one .4 The genetic distances among five breed are not very big (0.0303~0.2069), the smallest two populations are C.auratus yinji and C.auratus yiyu yinji (0.0303) ,which indicate the affiliation of such two population is the nearest ,instead ,the biggest two populations are C.auratus Yinji and C.auratus PengZiji(0.02069), which make us deduce the genetic varations between these two population is the biggest and the affiliation is farest . the cluster analysis results show that 5 crucians population are divide to three filiations ,one include C.auratus Yinji and C.auratus YiYu Yinji,the other one include C.auratus auratus and C.auratus PengZiji and C.auratus XiangYunji is independence.
