For scientific evaluation and protection scleractinian species resources, the researchusing microsatellite marker technology, microsatellite sequences were isolated fromPlatygyra acuta by using the magnetic bead hybridization enrichment method,18primerpairs were designed from microsatellite, then these sites primers were applied to amplifygenomic DNA in Platygyra acuta(30individuals), Platygyra daedalea(24individuals),Favites flexuosa(24individuals), Porites lutea(24individuals) four scleractinianpopulation genetic diversity. The results showed as follows:1. Microsatellite sequences were isolated from Platygyra acuta by using themagneticbead hybridization enrichment method.88positive clones were detected from328clones,18clones were discovered to contain microsatellites through sequencing analysis, Amongthe18microsatellites,12(66.67%)were perfect,3(16.67%) were imperfect and the rest3(16.67%)were compound type.2. To analyze genomic DNA diversity of four scleractinian population,18pairs ofprimers were designed from the microsatellite flanking sequences.10pairs of primers wereadapted to Platygyra acuta,11pairs of primers were adapted to Platygyra daedalea,9pairsof primers were adapted to Favites flexuosa,7pairs of primers were adapted to Poriteslutea.3. The results show the average polymorphism information content (PIC) are0.5168、0.4250、0.5286and0.3409;the average expected heterozygosity (He) are0.6667、0.6042、0.5972and0.4702; the average effective number of allele (Ne) are2.7112、2.5376、2.6860and2.0081. After comprehensive comparison inference: the degree of geneticpolymorphism in the Platygyra acuta and the Favites flexuosa population are high, thegenetic diversity of the Platygyra daedalea and the Porites lutea population wereat middle level.
