Purpose: Study the mechanism of tumor cells apoptosis induced by magnetic nanoparticles and magnetic nanoparticles modified with antisense oligodeoxynucleotide of human telomerase reverse transcriptase and the effect of P53 gene in this inducing process otherwise. Method: Atomic Force Microscope(AFM), Fluorescence microscope(FCM), transmission electron microscope(TEM) and flow cytometer and other apparatus also were used to test the occurrence of HL-60 tumor cell's apoptosis. We used Methyl Thiazolyl Tetrazolium(MTT), Wright's staining method and Colony Formation to observate the effect of growth inhibiting and cell multiplication of HL-60 tumor cell induced by magnetic nanoparticles and magnetic nanoparticles modified with antisense oligodeoxynucleotide of human telomerase reverse transcriptase. And we used Reverse Transcription-Polymerase Chain Reaction to detect the expression of P53 gene and the positioning of fluorescence Quantum Dots(QDs) in cells. We compared our magnetic nanoPolyAtract mRNA Isolation System with PolyAtract mRNA Isolation Systems of Promega as transverse experiement. This experiment clarifies application of magnetic nanoparticles to biology with different respects. Result 1 Magnetic nanoparticles, magnetic nanoparticles modified with antisense oligodeoxynucleotide of human telomerase reverse transcriptase induced HL-60 tumor cells to apoptosis, we could see typical morphologic change of apoptosis cells: karyopyknosis, chromation's condensing and aggregation in nuclear, forming crescent-shaped or annulus structures to lean on edge of cell nucleus's membrane and posing apoptosis body by Atomic Force Microscope (AFM), Fluorescence microscope (FCM), transmission electron Microscope (TEM) 2 There was a significant difference compared with control group(p<0.01), inhibition ratio had significant positive correlation with medication dosage and time ;during 0.8-8μM dosage amplitude, inhibition ratio accrescenced by dosage's increasing.
