The technique of insect cell culture, as one of the bases of cell engineering, is a valuable method of modern experimental biology. It is applied widely to various fields of biology, medicine and agriculture. Due to the situation that there is no any cell line from Coleoptera insects have been established at present in China, different tissues of two Coleoptera insects have been cultured in this study. It provided the necessary basic theory and useful reference for setting up the new insect cell lines from Coleoptera.And the chromosomes of ten insect cell lines also have been studied, which provided important theory reference for the research of insect cells and enriched the data of the insect cells bank. 1. Cell culture for two insects from ColeopteraIn this research, different tissues of Blaps rhynchoptera and Tenebrio molitor have been cultured in different culture conditions, and the suitable culture conditions of each tissue have been found out: (1) Cell culture for the neonate larvae of Blaps rhynchopter: Sterilized the surface of the eggs by 0.5% hibitane in 75% ethanol solution for 5 min could achieved the best sterilized effects and it wouldn't prevent the hatch of the eggs. The method of tissue fragment and modified Grace's medium were suitable for the cell culture of the neonate larvae of Blaps rhynchopter. (2) Cell culture for the embryos of Blaps rhynchopter: modified Grace's medium was more suitable for its culture. (3) Cell culture for the hemocytes of Blaps rhynchopter: It could achieved the best sterilized effects and the most amounts of blood by sterilized the adult insect in 75% ethanol for 8 min, and Schneider medium was favorable for its culture. (4) Cell culture for the testes of Blaps rhynchopter: It had no different results by the method of tissue fragment or the method of single cell, and Schneider medium was favorable for its culture. (5) Cell culture for the ovaries of Blaps rhynchopter: The method of single cell was better than the method of tissue fragment for its culture, and the results of four mediums heve been used in the culture were similar. (6) Cell culture for the fat bodies of Tenebrio molitor. It had no different results by the method of tissue fragment or the method of single cell, and Schneider medium was favorable for its culture.
