Optimize The Suspension Culture System Of Auxin-Habituated Cell Line Of Aralia Elata For Producting Useful Substances

In this paper,the accumulation of total flavonoids and oleanoli acid in the Aralia elata auxin-inhabituated cell line,suspended in the 250ml-Erlenmeyer flask with 100ml of medium,was studed with the methods of UV spectrophotometer and HPLC technology.Also,the kinds of nutrients in the culture mdium were analyzed.Those results were as follows:1.Different concents of sucrose(20--110 g/L)had a significant impact on cell growth and active substance accumulation.The highest growth of cells suspended culture for 21 d in the mdium with 90 g/L of sucrose,and their dry weigh is 5.17 times than those of others.The the highest contents of oleanolic acid in the cell was 1.78mg/g,which was obtained in the culture medium with 50 g/L of sucrose for 15 d;and in the same medium,the highest contents of total flavone in cells was 9.12 mg/g,cutlured for 18 d.2.The value of pH in the media with different sucrose contents during the cell suspent culture,also shew the decreasing first and then increasing,i.e it was decreased for the beginning 3 d,and then gradually increased after 3 d.during the cell suspension culture of 24 d,the value of pH of medium with 5%of sucrose always was higher than those others.The sucrose contents with the beginning of 5,7,9 and 11%after 24 d of culture,were 1.8?3.4?3.8?4.8%,respectively;among the ingrendients of the phosphate,the ammonium and the nitrate in the media,the depletion of phosphate by cells was the fastest after 12 d of culture.3.The dry weigh of cell cultured in the media with 1020 mg/L phosphates for 21d,was the highest value,among the 170-1360 mg/L of phosphate contents,0.91g.The content oleanolic acid in the cells cultured in the medium with the 680mg/L of phosphate for 21 d,was the highest,0.966mg/g;the content of total flavone in the cells cuture in the medium with the 340mg/L of phosphate for 18 d was the highest,5.49 mg/g.4.4 Sucrose consumption suspension cells during suspension culture in five phosphate concentrations were not significantly different,for the the culture of 24 d,the sucrose solution still 20g/L.After of the culture of 15d,the phosphate in the media with the contents of 170,340,680 mg/L were rapidly absorpted by cells,but there was still a few of it in the media with the 1020 and 1360 mg/L of it after the culture of 24d.The different contents of phosphates in media did not effect on the cell use for the ammonium and nitrate.
