There are studies about the tufted deer since 1980s and there is karyotype polymorphism in the species of the tufted deer. We had found 5 different karyotypes in the tufted deer as far as 2002. The chromosome which has much heterochromatin shows the main difference between different karyotype. There were different views about which chromosome is the sex chromosome. Besides, if the karyotype polymorphism in the species of tufted deer is the difference between subspecies or different individual, and the mechanisms of the polymorphism, and so on ,is still can't explan well. In order to check the sex chromosome of the tufted deer and discuss the questions above we have done such work:1) Y2 chromosome of M. m vaginalis is obtained by microdissection technology, and probes are made with DIG labels after the DNA copies by LA-PCR(linker-adaptor PCR)are amplified. Labeled probes are detected by Southern blot hybridization. The Sry gene was amplified using the genomic DNA of the tufted deer as template and made Sry gene probe by PCR with fluorescein after checked by sequencing. Then in situ hybridization of metaphase karyotype of male tufted deer are made use this two kind probes. The paint probe detected positive result. And we checked the Y chromosome of tufted deer associate with gene localization by our lab.2) Chromosomal analysis of skin cells from 8 tufted deer were studied by using primary culture and subculture.We found 4 different karyotypes in all and a new karyotype, different with the others studied before, was found. There were 47 chromosomes in somatic cell of the new karyotype, including a pair of heteromorphic X chromosomes, one was telocentric chromosome, the other was subtelocentric chromosome. So far, we have found 6 different karyotypes. We analyzed the pattern, heredity rule and possible mechanisms of karyotype polymorphism in the species of the tufted deer.3) Centromere probe was made by PCR with fluorescein. And then in situ...
