The Procapra przewalskii is an endangered species distributing only in the China. The conservation of the species' genetic research is the basis for the implementation of projects Procapra przewalskii rescue work. This essay explores using micosatellite DNAs to study the genetic diversity, and its major contents are as follows:The contrast among different methods of feces preservation: The methods are that feces was respectively stored in the field during collection at a low temperature after drying at 60 ℃, stored in 75% ethanol, stored in 100% ethanol and directly stored at a low temperature(in the attemperator added the biological ice bag),The result showed that DNA could be extracted successfully from the feces in a short storing period with any of these methods. What's more, the DNA sample stored in 100% ethanol could be longtime stored. At the same time, the modified DNA extracted method was a reliable method to obtain high quality DNA from the Procapra przewalskii feces samples. It established a convenient method to save the collectes feces in the field and transport back to the laboratory for DNA analysis. And we could use these methods in other endangered species for their genetic studies.The analysis of genetic diversity of the HaErgai population: 12 high polymorphic microsatellite loci were screened from 40 microsatellite loci derived from cattle and sheep by PCR in 43 Procapra przewalskii feces samples which were collected at HaErgai area in Qinghai Province. In these 12 loci, 94 alleles were found and the effective number of allele (Ne) ranged from 9.3858 to 4.3867. The polymorphism information content (PIC) value at twelve microsatellite ranged from 0.8841 to 0.8026. The average number of genetic heterozygousity (He) is 0.8483. The results revealed that the HaErgai population took on a high level of genetic diversity.The analysis of genetic diversity of the main populations: the present study was presented on 12 microsatellite loci analyses of six populations samples collected from TianJun, HaErgai, Bird Island, the railway station of Qinghai lake, YuanZhe and HuDong. The results showed that there was a long divergent times between TianJun, and Bird Island, Genetic distance between the HaErgai population and YuanZhe population is the smallest. The UPGMA cluster analysis clustered the black line of YuanZhe population and the HuDong population line were clustered at first, then the black line of HaErgai population and the railway station of Qinghai lake population line, but TianJun and Bird Island are single population.
