From the beginning of the RAPD.the most problem embarrass the reaches is the instability of this technology.People found the mismatch not tar away from this technology burn.Elena took a series experiments of RAPD to study the kind of the mismatch. They found that the mismatch happen very frequency.In our experiment we according tO the theory,of Nearest - Neighbor.combined the data of SantaLucia's years work.We take the flee energy as a stand of the mismatch in RAPD and we statistic the value of all kind of mismatch.By this data we find the area of the value of free energy.During the area, the site of mismatch still can amplified the profile of RAPD.but the value under these rang the amplified instability.And for the development research the cool wheat.we built a stable system for RAPD in wheat.The main results were as follows:1.Find the primer which can produce different profile between the cold and warm parent.2. The result of analyzed by theory for RAPD technology,there were a lots of mismatch during the PCR of RAPD3.By use of the Gibbs free energy to calculate the combine energy,we can statistic the mismatch in RAPD more accurately.
